高效液相色谱流动相添加剂法拆分亚叶酸钙对映体
Separation of Enantiomers of Calcium Folinate by HPLC with Chiral Mobile Phase Additives
【摘要】 目的:利用高效液相色谱手性流动相添加剂法拆分亚叶酸钙对映异构体。方法:通过探讨色谱柱、柱温、手性配合剂、流动相的pH值对手性拆分的影响,确定乙腈-苯丙氨酸溶液(8 mmol L-苯丙氨酸、4 mmol硫酸铜,加水1000 m L,氢氧化钠试液调节pH至3.5)(10∶90)为手性流动相,Kromasil C18柱(250 mm×4.6 mm,5μm)拆分亚叶酸钙对映体,检测波长为300 nm,柱温20℃。结果与结论:建立的方法简单易行,可用于药品质量控制与体内代谢立体选择性的研究。
【Abstract】 Objective: To establish a method to separate the enantiomers of calcium folinate by HPLC with chiral mobile phase additives. Methods: Factors affecting chiral separating, including pH of mobile phase, chiral additives, column temperature and stationary phases, were optimized. The separation of the enantiomers was established on a kromasil C18column(250 mm×4.6 mm, 5 μm) at 20 ℃ with acetonitrilephenylalanine solution(containing 8 mmol·L-1phenylalanine and 4 mol·L-1 copper sulphate, adjusted to pH=3.5)(10:90) as chiral mobile phase and detected at 300 nm. Results and Conclusion: The proposed method is simple and can be used for the quality control of calcium folinate enantiomers.
