TNS与人中心蛋白2作用的荧光光谱研究
Characterization of TNS Binding with Human Centrin 2
【摘要】 在0.01mol/L Hepes,pH 7.4及室温条件下,采用荧光光谱研究了探针2-对甲苯胺基-6-萘磺酸(2-p-toluidinylnaphthalene-6-sulfonate,TNS)与人中心蛋白2(human centrin 2,HsCen2)的作用。结果表明,与HsCen2结合后TNS荧光峰位置发生蓝移,荧光强度明显增大;440nm处滴定曲线表明HsCen2与TNS的结合比为1∶1;通过Frster无辐射能量转移原理,计算得到TNS与HsCen2中Tyr172的距离为r=2.13nm;疏水性相互作用是TNS与HsCen2作用的主要驱动力。
【Abstract】 The binding of hydrophobic fluorescent probe TNS with human centrin 2(HsCen2)was examined by fluorescence spectra in 0.01mol/L Hepes,pH 7.4.While combined with HsCen2,the fluorescence peak position of TNS was blue-shifted and the fluorescence intensity was increased to 4.8times.HsCen2 could combine one TNS molecule by hydrophobic interaction.According to Frster non-radiative energy transfer,the distance between HsCen2Tyr172 and TNS was calculated to be 2.13 nm.
