结直肠癌组织中miR-150表达及其对癌细胞增殖的影响
Expressions of miR-150 in colorectal cancer and its influence on cell proliferation
【摘要】 目的观察结直肠癌组织中miR-150的表达变化,探讨miR-150对结直肠癌细胞增殖的影响。方法选择35例结直肠癌患者,采用实时荧光定量聚合酶链反应(qRT-PCR)检测癌组织和癌旁正常组织中miR-150的表达。将结直肠癌HCT116细胞分为两组,分别采用miR-150模拟物(转染组)和阴性对照(对照组)进行转染,采用CCK-8法检测两组的细胞增殖活性。结果 miR-150在结直肠癌组织中的相对表达量为0.071(0.031,0.129),癌旁正常组织中为0.336(0.168,0.817),癌组织明显低于癌旁正常组织(P<0.05)。与对照组比较,转染组转染48、72、96、120 h时细胞增殖活性显著降低(P均<0.05)。结论结直肠癌组织中miR-150表达下调;miR-150具有抑制结直肠癌细胞增殖的作用,上调miR-150表达可抑制结直肠癌细胞增殖。
【Abstract】 Objective To observe the expression changes of miR-150 in colorectal cancer tissues and to investigate the influence of miR-150 on cell proliferation of colorectal cancer. Methods The expression of miR-150 in 35 cases of colorectal cancer tissues and adjacent normal tissues was detected with real-time fluorescent quantitative polymerase chain reaction( qRT-PCR). HCT116 cell line was transfected with miR-150 mimic and negative control respectively,and the cell proliferation activities of the two groups were detected with CCK-8 method. Results The expression of miR-150 in the colorectal cancer tissues and adjacent normal tissues was 0. 071( 0. 031,0. 031) and 0. 336( 0. 168,0. 168),respectively. The expression of miR-150 in the colorectal cancer tissues was significantly lower than that of the adjacent normal tissues( P < 0. 05). Compared with the control group,the activity of HCT116 cell proliferation in the transfection group was significantly decreased at 48 h,72 h,96 h and 120 h( all P < 0. 05). Conclusions The expression of miR-150 in the colorectal cancer tissues is down-regulated. The miR-150 has inhibitory effect on colorectal cancer and the up-regulated expression of miR-150 can inhibit the proliferation of colorectal cancer cells.
