红景天提取液对大鼠心肌缺血预处理的实验研究

【摘要】 目的探讨红景天提取液对大鼠心肌缺血预处理后的心肌保护作用机制。方法将20只健康大鼠随机分为模型组、红景天组,两组均经尾静脉注射垂体后叶素,建立急性心肌缺血模型,红景天组尾静脉注射红景天提取液2 mL/(kg·d),模型组给予等体积生理盐水。于缺血前及缺血预处理1周后,尾静脉取血测定血清超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性,断头取心,以ELISA法测定大鼠缺血心肌组织受体胎肝激酶1(FLK-1)蛋白及FLK-1a mRNA,缺氧诱导因子-1a(HIF-1a)蛋白及HIF-1a mRNA,缺血心肌组织中丙二醛(MDA)。结果与模型组及缺血预处理前比较,红景天组在缺血预处理1周后FLK-1蛋白及FLK-1 mRNA、HIF-1a蛋白及HIF-1a mRNA表达明显增强,CAT、SOD活性提高,MDA含量明显降低,差异均有统计学意义(P<0.05)。结论红景天提取液可提高预处理后FLK-1蛋白、HIF-1a蛋白、FLK-1 mRNA、HIF-1a mRNA表达及自由基清除酶活性,减轻急性心肌缺血大鼠血管内皮炎症反应,抑制脂质过氧化反应,对心肌缺血有一定的保护作用。

【Abstract】 Objective To investigate the protective mechanism of rhodiola liquor in myocardial ischemic preconditioning in rats. Methods Twenty healthy rats were randomly divided into model group and rhodiola group. The acute myocardial ischemia model was established with injection of posterior pituitary hormone through caudal vein. The rhodiola group was injected with rhodiola liquor [2 mL /( kg·d) ] and the model group with saline [2 mL /( kg·d))through caudal vein. The blood was gathered before heart ischemia and one week after heart ischemia. The protein and mRNA were extracted from myocardium. The activity of SOD and CAT was measured. The expression of HIF-1a,FLK-1,HIF-1a mRNA,FLK-1 mRNA and MDA was measured. Results Compared with the model group and the rhodiola group before myocardial ischemic preconditioning, the expression of FLK-1,FLK-1 mRNA,HIF and HIF-1a mRNA were significantly increased in the rhodiola group. The activity of SOD and CAT was significantly enhanced. However,the content of MDA was significantly decreased( P < 0. 05). Conclusion The rhodiola liquor can increase the activity of free radical eliminate enzyme,FLK-1 protein, and HIF-1a protein, increase the expression of FLK-1 mRNA and HIF-1a mRNA, reduce acute inflammation reaction of vascular endothelium,and inhibit the lipid peroxidation,and it has protective effect on myocardial ischemia.
