紫草素对银屑病样动物模型Th17细胞相关因子的影响
Effects of Shikonin on Th17 related cytokines in psoriasis-like mouse model
【摘要】 目的:探讨紫草素对银屑病样动物模型Th17细胞相关因子的影响。方法:将6~8周龄雌性BALB/c小鼠随机分为空白对照组、低剂量紫草素组、高剂量紫草素组、甲氨蝶呤(MTX)组及模型组。采用银屑病皮损面积及严重程度指数(PASI)作为评分标准,记录小鼠皮损动态变化;光镜观察皮损组织形态学变化,并测量表皮层厚度;酶联免疫吸附试验(ELISA)检测小鼠血清Th17细胞相关因子浓度变化。结果:咪喹莫特(IMQ)处理的小鼠背部皮肤出现典型的红斑、鳞屑及皮肤增厚等银屑病样皮损,随时间增加,PASI评分表现为先增加后降低的趋势。低、高剂量紫草素组及MTX组PASI评分较模型组低;皮损组织病理示模型组表现为显著的角化过度伴角化不全,棘层明显增厚;与模型组相比,低、高剂量紫草素组及MTX组表皮层厚度明显低(P<0.01)。ELISA结果示低、高剂量紫草素组及MTX组小鼠血清中Th17细胞相关因子白介素(IL)-17、IL-17F、肿瘤坏死因子(TNF-α)、IL-6、IL-22等水平均低于模型组(P均<0.05)。结论:紫草素可明显阻断银屑病样皮损及皮损组织病理变化的出现,并降低血清中Th17细胞相关因子水平。
【Abstract】 Objective: To investigate the effects of shikonin on Th17 related cytokines in psoriasis-like mouse model.Methods: Six to eight-week-old female BALB/c mice were randomly divided into blank controls group, low-dose shikonin,high-dose shikonin, MTX and untreated psoriasis-like mice. Psoriasis area and severity index(PASI) score was used to assess changes in skin lesions. Light microscope was used to evaluate morphological changes and to measure the epidermal thickness. The levels of serum cytokines were detected by ELISA. Results: Imiquimod treatment induced typical psoriasis-like skin lesions such as erythema, scales, and thickening. PASI scores increased gradually, followed by a gradual decline over the course. PASI scores were improved after treatments with either shikonin or MTX. Imiquimod-induced morphological changes included epidermal hyperkeratosis, parakeratosis, and acanthosis. Treatments with either shikonin or MTX significantly reduced epidermal thickness(P<0.01). Similarly, treatments with either shikonin or MTX significantly lowered serum levels of IL-17、IL-17F、TNFα、IL-6 and IL-22(all P<0.05, vs. untreated psoriasis-like mice). Conclusion:Shikonin could significantly improve skin lesions and lowered the serum levels of Th17 related cytokines in psoriasis-like mouse model.
