珍稀濒危植物异形玉叶金花愈伤组织的诱导
Callus Induction of Mussaenda anomala,a Rare and Endangered Plant
【摘要】 为珍稀濒危植物异形玉叶金花种质资源的繁殖、保存与利用提供依据,以幼嫩叶片为外植体,采用L16(45)正交试验设计研究不同浓度2,4-D、6-BA、暗培养时间及叶片划痕数等因子对异形玉叶金花叶片诱导愈伤组织的影响。结果表明:各因素对愈伤组织诱导率的影响依次为2,4-D>暗培养时间>6-BA>划痕数;异形玉叶金花叶片诱导愈伤组织各因素最佳组合为1.5mg/L 2,4-D+0.3mg/L 6-BA,划痕数5~7条,暗培养20d。该条件下愈伤组织的诱导率达91.6%。
【Abstract】 The effect of 2,4-D,6-BA,dark culture days and scratch number on callus induction of young M.anomala leaves by L16(45)orthogonal test design to provide the basis for reproduction,conservation and utilization of rare and endangered M.anomalagermplasm resources.Results:The effect degree of four factors to influence callus induction rate is 2,4-D > dark culture days > 6-BA > scratch number.The optimal combination of four influence factors for callus induction of young M.anomalaleaves includes 1.5mg/L 2,4-D,0.3mg/L 6-BA,5~7scratches and dark culture for 20 d.Under this condition,the induction rate of callus was 91.6%.
