不同浓度骨髓间充质干细胞对肺气肿大鼠模型的影响

【摘要】 目的观察骨髓间充质干细胞(BMSC)对肺气肿模型大鼠炎症因子和肺组织病理学变化的影响,探讨肺气肿的发病机制。方法采集3只SD幼鼠骨髓细胞,培养传代;将40只雄性SD大鼠随机分成对照、肺气肿模型组、高、中、低浓度BMSC组,建立肺气肿模型,大鼠尾静脉注射不同浓度的BMSC治疗。观察肺组织病理学变化及肺泡灌洗液细胞计数,用ELISA检测各组大鼠BALF中IL-17和IL-6水平。结果肺气肿模型组和3个BMSC组BALF中白细胞总数、巨噬细胞数及中性粒细胞百分比均高于对照组(P<0.05)。与正常组比较,3个BMSC组、肺气肿模型组BALF和血清中IL-17、IL-6水平明显升高(P<0.05)。BMSC组、肺气肿模型组与正常组比较,MLI增加,MAN减少(P<0.05)。结论BMSC能改善肺气肿大鼠模型的病理改变,且浓度越高疗效越好。

【Abstract】 Objective To investigate the effects of bone marrow mesenchymal stem cells( BMSCs) of emphysema in rats inflammatory cytokines in lung tissue and pathological changes so as to investigate the pathogenesis of emphysema. Methods Three SD rats were collected in the bone marrow cells suspension. Forty male SD rats were randomly divided into normal control group,emphysema model group,high,middle and low concentration BMSC groups. The emphysema model was established,and the rats were treated with different concentrations of BMSC in the tail vein injection. The pathological changes of lung tissue and alveolar lavage cells were observed,and the levels of IL- 17 and IL- 6 in BALF were detected by ELISA method. Results The emphysema model group and three groups of BMSC in BALF in total white cell count,the number of macrophages and neutrophil percentage were higher than in the normal control group( P < 0. 05); Compared with the three groups of BMSC group,emphysema model group compared with normal group,in BALF and serum levels of IL- 17 and IL- 6 increased significantly( P < 0. 05); Compared with normal group,the MLI increased and the MAN reduced in BMSC group and the emphysema model group( P < 0. 05). Conclusion Intravenous injection of BMSC can improve the pathological changes of the rat model of emphysema,and the higher the concentration,the better effect.
