硼替佐米对Raji细胞增殖及SHP-1、c-kit基因表达的影响

【摘要】 目的:本实验拟从细胞生物学行为的角度,探讨硼替佐米对Raji细胞株的影响。方法:用细胞培养的方法,传代培养Raji细胞株,用不同浓度的硼替佐米,分别对Raji细胞进行干预,并于干预后不同时间收取各组细胞。细胞增殖抑制率通过MTT法来检测,应用药物前后细胞中SHP-1基因、c-kit基因的表达情况通过实时定量PCR的方法来检测。结果:(1)硼替佐米可明显抑制Raji细胞的增殖,而且增殖抑制率与药物的浓度及作用时间呈正相关。(2)Raji细胞受硼替佐米干预后,细胞中SHP-1基因表达水平增加;c-kit基因表达水平降低。结论:硼替佐米可以抑制Raji细胞增殖,硼替佐米诱导Raji细胞凋亡的过程,可能受到SHP-1基因及c-kit等多种相关基因的共同调控。

【Abstract】 Objective: This study was aimed to explore the growth inhibition of Bortezomib on lymphoma celllines Raji cells.Methods:Human lymphoma cells Raji were cultured and treated with Bortezomib of different doses.Wedetect inhibition rates of the Raji cells with MTT.Real-time quantitative PCR was used to detect SHP-1, c-kit mRNA expression.Results:(1)Bortezomib can inhibit Raji cells,and the effect was related to the concentration and acting time of Bortezomib.(2)The expression level of SHP-1mRNA after bortezomib in Raji cells was increased.The expression of c-kit mRNA was decreased.Conclusion:Bortezomib can inhibit Raji cells proliferation.SHP-1 and various relateted gene might together participate in the induced apoptosis of Raji cells with Bortezomib.
