“一锅法”合成PCL-b-PEG嵌段聚合物及其自组装行为

【摘要】 利用点击化学与酶促开环聚合相结合的"一锅法"制备聚己内酯-b-聚乙二醇(PCLb-PEG)两亲性嵌段聚合物.首先通过对二氯代乙氧基乙醇进行叠氮化亲核取代修饰得到了同时引发点击化学反应与酶促开环聚合反应的双官能引发剂;再利用戊炔酸对聚乙二醇炔基化修饰;最后由双官能引发剂的叠氮端基引发炔基化PEG的点击化学,同时在生物催化剂Novozyme-435的催化下利用其羟端基引发己内酯的酶促开环聚合.经核磁共振与GPC的测试对所制备嵌段聚合物结构进行了表征并验证了一锅法合成的可行性.由GPC测试得到相对分子质量为15 093、多分散性良好(1.18)的嵌段聚合物.经过动态光散射测试得到水粒半径为202nm,而由透射电子显微镜测试所得的干燥状态下粒子半径为146nm.

【Abstract】 Biodegradable and biocompatible PCL-b-PEG amphiphilic block copolymers were prepared by combination of Enzymatic ring-opening polymerization and "click"chemistry via "one-pot"method under mild conditions.First the bifunctional initiator was prepared by chlorinated ethoxy ehanol which was displaced by azide nucleophilic and additionaly the polyethyleneglycol was modified by utilized pentynoic acid.Ultimately,the bifunctional initiator initiated click chemistry and enzymatic ringopening polymerization in the present of biocatalyst Novozyme-435.Click chemistry of the alkynylation PEG was initiated by azido-ended of the bifunctional initiator,simultaneously enzymaic ring-opening polymerization was triggered by the other hydroxyl-ended with the present of biocatalyst Novozyme-435.The composition and block architecture of the copolymers were characterized by 1 H NMR,FTIR and GPC analyses.These amphiphilic block copolymers could self-assemble into sphere-like aggregates in aqueous solution with diverse diameters.The size and shape of the block-copolymers micelles were investigated by using dynamic light scattering(DLS)and transmission electron microscope(TEM).The results of DLS and TEM indicate that the copolymers micelles appear a spherical shape and had a mean diameter(146nm).
