佳宝苦瓜杂交种纯度的SRAP鉴定

【摘要】 利用SRAP技术对佳宝苦瓜杂交种子进行纯度鉴定。试验结果表明,从153对SRAP引物中筛选出1对引物Me6-Em4,能同时扩增出F1代及其父母本的多态性条带,且PCR扩增产物具有高度的稳定性和重复性;利用该引物对48株佳宝苦瓜进行纯度鉴定,其种子纯度为97.9%,与其田间鉴定结果接近,表明利用SRAP分子标记可以快速、准确地鉴定佳宝苦瓜杂交种纯度。

【Abstract】 In this study,SRAP markers technique was used for identifying the purity of Jiabao bitter gourd hybrid seeds.The results showed that only a pair of primers(Me6-Em4) was screened out from 153 pairs of SRAP primers,which could amplify the characteristic polymorphic bands of F1 generation and its parents,and the PCR products possessed high stability and repeatability.Using the pair of primers to detect the purity of 48 individuals of Jiabao bitter gourd,the results showed that the purity was 97.9% for hybrid seeds of Jiabao bitter gourd,which was very close to the field morphological test,indicating that SRAP markers could be used to identify the hybrid purity of Jiabao bitter gourd rapidly and accurately.
