IGF-1基因多态性与猪生长性状的相关性分析

【摘要】 目的:研究淮猪新品系与杜洛克猪杂交的生产性能,并分析IGF-1基因对猪生长性状的遗传效应。方法:通过ACEMA-64自动饲喂系统测定杜洛克猪、淮猪新品系、杜洛克猪×淮猪新品系的生长性状。另外,用PCR-RFLP方法对测定猪的基因多态性进行检测。结果:杜洛克猪、淮猪新品系×杜洛克猪的平均日增重和达100kg校正日龄都显著优于淮猪新品系(P<0.05);淮猪新品系×杜洛克猪的饲料转化率显著低于淮猪新品系(P<0.05),但也显著高于杜洛克猪(P<0.05)。IGF-1基因PCR产物经Hha I酶切,杜洛克猪仅检测到BB型;淮猪新品系检测到AB和BB型;杜洛克猪×淮猪新品系检测到AA、AB和BB型。关联分析表明,三种基因型个体的平均日增重、饲料转化率和达100kg校正日龄存在差别,但是差异都不显著(P>0.05)。其中,淮猪新品系BB型个体的生长性状有优于AB型的趋势,杜洛克猪×淮猪新品系BB型个体有优于AB和AA型的趋势。结论:淮猪新品系与杜洛克猪杂交效果较好,但IGF基因作为改善猪生长性状的分子标记仍需进一步的验证。

【Abstract】 Objective: To explore the production performance of Huai pig hybrided with Duroc pigs,and to analyze the genetic effect of the IGF- 1gene on growth traits in pigs. Method: Growth traits of Duroc pigs,Huai pig and Duroc pigs × Huai pig were measured by the automatic feeding system of ACEMA64. In addition,the IGF- 1 gene polymorphism was detected by polymerase chain reaction- restriction fragment length polymorphism( PCR- RFLP) method in these tested pigs. Results: The average daily gain and corrected day at 100 kg of Duroc pigs and Duroc pigs × Huai pig were significantly better than Duroc pigs × Huai pig( P < 0. 05); the feed conversion rate of Duroc pigs × Huai pig was significantly lower than Huai pig( P < 0. 05),but was significantly higher than Duroc pigs( P < 0. 05). PCR products of the IGF- 1 gene were digested with the Hha I enzyme,and only BB genotype was detected in Duroc pigs; AB and BB genotypes were detected in Huai pig; AA,AB and BB genotypes were detected in Duroc pigs × Huai pig. Association analysis showed that the differences were existedin the average daily gain,feed conversion rate,and corrected day at 100 kg among three genotype individuals,but the differences were not significant( P > 0. 05). Among them,the growth traits of individuals with the BB genotype had a better trend than that of AB genotype in Huai pig new line,and individuals with the BB genotype had a better trend than that of AB and AA genotypes in Duroc pigs × Huai pig new line. Conclusion: Huai pig new line and Duroc pigs have a good hybrid effect,but the IGF- 1 gene is taken as a molecular marker for improving the growth traits of pigs still needs further verification.
