不同抗生素对大鼠上呼吸道菌群定植影响的研究
Different antibiotic-driven changes in upper airway microbiota colonization in rats
【摘要】 目的研究不同种类抗生素对大鼠上呼吸道菌群定植的影响,为探讨菌群定植对呼吸道免疫耐受形成的影响提供合适的动物模型。方法将Wistar大鼠分为阴性对照组(N)、阿奇雾化组(A)、头孢雾化组(T)、磷霉素雾化组(P)。采用雾化抗生素的方法,让Wistar幼鼠吸入抗生素,1周后将大鼠处死,取鼻腔灌洗液,DGGE分析上呼吸道菌群变化。结果三组抗生素处理组DGGE条带数均少于正常组,其中头孢噻肟组上呼吸道灌洗液菌群DGGE条带数明显少于正常组(P<0.05),且绿色气球菌、类鸟肠球菌、阴沟肠杆菌含量增加。结论大鼠幼年使用抗生素会改变上呼吸菌群的正常定植,头孢雾化可以得到合适的呼吸道菌群紊乱模型。
【Abstract】 Objective To study the influence of different antibiotics on the airway microbiota,and provide an appropriate animal model to demonstrate the effect of normal microbiota on the maturing of immune tolerance. Methods Wistar rats were randomly divided into Normal group,Azithromycin group,Cephalosporin group,and Fosfomycin group. 1 week after neonatal Wistar rats inhaled the antibiotics,the animals were sacrificed with intraperitoneal injection of 10% chloral hydrate; nasal lavage fluids were obtained,and the changes of upper airway microbiota were measured by using DGGE. Results The airway microbiota in cephalosporin group reduced significantly compared with that in normal group( P < 0. 05),while specific bacterial bands such as Aerococcus viridans,Enterococcus pseudoavium and Enterobacter cloacae increased. Conclusion Antibiotic treatment has a significant influence on the colonization of airway normal microbiota of neonatal rats,and cephalosporin inhalation can build an appropriate model of airway microbiota dysbiosis.
