唾液乳杆菌FDB86小试高密度发酵培养基的优化研究
Optimization of Culture Medium of Lactobacillus salivarius FDB86
【摘要】 唾液乳杆菌FDB86是一株具有优良功能的益生菌。为了实现菌株的产业化应用,本研究对该菌株小试高密度发酵培养基进行了优化。以唾液乳杆菌FDB86的活菌浓度为评价指标,以MRS培养基为基础培养基,利用正交试验的方法,对培养基中碳源、氮源以及无机盐添加量进行了优化,确定活菌浓度最佳的培养基配方为:蛋白胨5.0g/L,酵母粉7.5g/L,牛肉膏10.0g/L,葡萄糖15.0g/L,无水乙酸钠8.0g/L,七水合硫酸镁1.0g/L,四水合硫酸锰0.1g/L,柠檬酸氢二铵2.0g/L,磷酸二氢钾2.0g/L,吐温-80 1.0g/L。对优化完成的培养基进行50L发酵罐小试试验,结果表明唾液乳杆菌FDB86约在6h达到稳定期,菌体浓度可以达到7.5×109CFU/m L,与基础培养基培养结果(2.4×109CFU/m L)相比有显著的提高,具有实际应用价值。
【Abstract】 The strain for this study is Lactobacillus salivarius FDB86, with MRS culture medium based, using orthogonal test method, viable count as the evaluation index, to optimize adding quantity of cultivate medium carbon source, nitrogen source and inorganic salt. Final determine the viable count of the best culture medium formula: Peptone 5.0g/L, Yeast Extract Powder 7.5g/L, beef extract 10.0g/L, D-Glucose15.0g/L, Sodium acetate anhydrous 8.0g/L, Magnesium sulfate heptahydrate1.0g/L, Manganese sulfate monohydrate 0.1g/L, di-Ammonium hydrogen citrate 2.0g/L, di-Potassium hydrogen phosphate trihydrate 2.0g/L,Tween 80 1.0g/L.Through 50 L fermenter smallscale test to examine the effect of optimized culture. The results show that Lactobacillus salivarius FDB86 about at 6h reached stationary phase, cell concentration reached 7.5x109CFU/m L, showed that the optimized culture effect is better, has the value of practical application.
