早产大鼠胎盘白细胞介素-6、白细胞介素-10水平与支气管肺发育不良的相关性
Correlation between IL- 6,IL- 10 levels in placenta of premature rats and bronchopulmonary dysplasia
【摘要】 目的探讨早产大鼠胎盘白细胞介素-6(IL-6)、白细胞介素-10(IL-10)水平与支气管肺发育不良(BPD)的相关性。方法选择定期受孕的Sprague Dawley(SD)大鼠,于孕第20天剖宫产取出胎鼠及胎盘,其胎盘组织内IL-6、IL-10均采用免疫组织化学法及Western blot法(免疫印迹法)进行检测。早产鼠出生后置于高氧中持续喂养,于高氧暴露第14天,取早产鼠肺用于组织切片,进行苏木精-伊红(HE)染色后,观察肺组织形态学结构及放射性肺泡计数,从而判断早产鼠是否发生了BPD。根据肺泡计数及肺泡结构分为BPD组和非BPD组,分析两组胎盘IL-6、IL-10水平与早产鼠发生BPD的关联性。结果 BPD组胎盘IL-6表达水平明显高于非BPD组,差异有统计学意义(P<0.05);非BPD组胎盘IL-10表达水平高于BPD组,差异有统计学意义(P<0.05)。结论 IL-6可能促进BPD的发生,IL-10可能减少BPD的发生。
【Abstract】 Objective To explore the correlation between interleukin- 6( IL- 6),interleukin- 10( IL- 10) levels in placenta of premature rats and bronchopulmonary dysplasia( BPD). Methods SD rats of regular conception were selected,preterm rats and placenta specimens were obtained by cesarean section on the 20 th day of gestation. The expression levels of IL- 6 and IL- 10 in placenta were detected by immunohistochemistry and Western blot. Preterm rats were fed in highly oxygenated environment after birth. On the 14 th day after high oxygen exposure,lung pathological changes of the preterm rats were observed by HE staining and radical alveolar counter. The preterm rats were divided into BPD group and non- BPD group according to morphological structure of lung tissue and radical alveolar counter.The correlations between IL- 6,IL- 10 levels in placenta of premature rats and BPD of preterm rats were analyzed. Results The expression level of placental IL- 6 in BPD group was statistically significantly higher than that in non- BPD group( P < 0. 05); the expression level of placental IL- 10 in non- BPD group was statistically significantly higher than that in BPD group( P < 0. 05). Conclusion IL- 6may promote the occurrence of BPD,IL- 10 may reduce the occurrence of BPD.
