氧化苦参碱对异丙肾上腺素致心肌肥厚大鼠心脏重构的保护作用研究

【摘要】 目的:研究氧化苦参碱对异丙肾上腺素致心肌肥厚大鼠心脏重构的保护作用,并探讨其作用机制。方法:给予Spraguedawley大鼠连续7 d皮下注射异丙肾上腺素5 mg/(kg·d),以建立大鼠心肌肥厚模型,观察氧化苦参碱对模型大鼠体质量,心肌肥厚指数,心肌组织形态学改变,超氧化物歧化酶、丙二醛及血清一氧化氮含量,一氧化氮合酶活性的影响。结果:氧化苦参碱可明显降低模型大鼠心肌肥厚指数、心肌细胞直径与横截面积、心肌间质胶原纤维含量;增强了血清及心肌组织中超氧化物歧化酶的活力,并相应地降低了脂质过氧化物丙二醛的含量;增强了血清中总一氧化氮合酶的活性,增加了血清一氧化氮的含量,但降低了诱导型一氧化氮合酶的活性。结论:氧化苦参碱对异丙肾上腺素致心肌肥厚大鼠有明显的心脏重构保护作用,其作用机制可能与提高机体抗氧化酶活性和调节血清一氧化氮含量/一氧化氮合酶活性有关。

【Abstract】 OBJECTIVE: To investigate the protective effect of oxymatrine on cardiac remodeling in rats with isoproterenol( ISO)-induced heart failure and explore its action mechanism. METHODS: Myocardial hypertrophy model was established in Sprague-Dawley rats by subcutaneous injection with isoproterenol[ISO,5 mg /( kg·d) ] for seven days. The body weight,myocardial hypertrophy indexes and histomorphology were followed to evaluate the effect of oxymatrine on cardiac remodeling in rats with isoproterenol-induced myocardial hypertrophy. In addition,levels of superoxide dismutase( SOD),malondialdehyde and serum nitric oxide( NO) and the activity of NO synthase( NOS) in serum were measured. RESULTS: Oxymatine treatment significantly decreased the myocaridial hypertrophy indexes,cardiomyocyte diameter and the content of cardiac interstitial collagen fiber,enhanced the activity of superoxide dismutase( SOD) in both serum and cardiac tissue and accordingly decreased the content of lipid peroxide malondialdehyde,enhanced the activity of serum NOS and serum content of NO but decreased the activity of NitricOxide Synthase of the induced type. CONCLUTIONS: The mechanisms underlying the significant protective effect of oxymatine against cardiac remodeling might be attributed to its action in enhancing the activity of antioxidase and regulating NO content and NOS activity.
