人脑胶质瘤干祖细胞-绿色荧光裸小鼠皮下移植瘤SU3-NC/GFP-1模型建立

【摘要】 目的探讨人脑胶质瘤干祖细胞-绿色荧光裸小鼠皮下移植瘤模型的建立。方法将人脑胶质瘤干祖细胞株SU3接种于NC-C57BL/6J荧光裸小鼠右前肢腋下,定期测肿瘤大小,取移植瘤组织块持续传代,并做常规病理、免疫组化染色和绿色荧光蛋白(GFP)检测。结果移植瘤连续传至10代,100%致瘤率。肿瘤晚期都有肉眼可见的肿瘤表面皮肤溃疡性结痂,常规病理符合恶性肿瘤特征,分子免疫病理缺氧诱导因子1α(HIF-1α)、CD34、TNF、Ki67非均一性表达于相关区域或相关细胞,在470nm激发光下可见移植瘤间质存在一定数量的GFP细胞,以及在坏死区域内的GFP细胞CD68+和CD11b+。结论成功建立人脑胶质瘤干祖细胞-绿色荧光裸小鼠皮下移植瘤模型。

【Abstract】 Objective To establish the GFP nude mice model with human glioma stem cell SU3-NC/GFP-1subcutaneous xenograft.Methods Human glioma stem cell line SU-3was inoculated subcutaneously into the transgenic NC-C57BL/6Jnude mice with whole body green fluorescence protein expression.Tumor size was measured in the living tumor-bearing mice at regular intervals.Then the transplantable tumor was harvested for routine pathological examination and immunohistochemical and GFP analysis.Results The transplantable tumor was passaged to the 10 th generation in vivo.The tumorigeneity rate was 100% without spontaneous remission.In the late stage,ulcer scar in the skin surface of the tumor-bearing mice could be observed.Conventional pathological analysis showed the characteristics of malignant tumor.Immunopathological results showed non-hemogeneous expressions of HIF-1α,CD34,TNF,Ki67 in tumor invading areas.The GFP cells could be observed in the tumor interstitial tissue under 470 nm excitation light,and CD68+,and CD11b+in necrotic area.Conclusion The fluorescent nude mice tumor model with human glioma stem cell SU3-NC/GFP-1subcutaneous xenograft has been established successfully.
