四种天然添加物对白及组培苗诱导的影响
On Evaluation of Seedling Potential Growth in Bletilla striata(Thunb.)Reichb.f.,Suffered from Four Kind of Natural Additives
【摘要】 白及是现代医药工业和化妆品工业的重要原材料,在基本培养基中添加不同比例的马铃薯、西红柿、香蕉和椰汁,检测种子萌发率和组培苗不同器官的表型数据,比较了不同天然添加物对白及组培苗生长势的影响,研究促进白及生长发育的营养参数.发现添加15%马铃薯利于白及叶片和茎的生长;添加20%的西红柿利于叶片生长,但对根的生长具有一定抑制作用;添加2%的香蕉显著促进白及块茎生长;添加1%的椰汁利于白及根的增殖;15%的马铃薯和1%的椰汁能促进白及植株的快速生长.白及种子在MS+2.0mg/L IBA+30.0g/L蔗糖+7.0g/L琼脂+15%马铃薯培养基上进行萌发,30d后继代到MS+2.0mg/L IBA+30.0g/L蔗糖+7.0g/L琼脂+1%椰汁培养基上培养,3个月后可以发育成茎粗、根多、苗壮的优质种苗.
【Abstract】 B.striatais an important raw material for modern industries of pharmaceutical and cosmetics in Chinese traditional medicine.To explore the parameters of produce and development,diverse natural additives have been applied to observe the seedling potential growth in B.striatathrough tissue culture technique.Phenotypic data of growth have been measured in the present experimental materials,depending on MS solution and diverse natural additives of tomato,potato,banana and coconut juice.Rapid growths of leaf and stem are observed in MS solution with 15% potato.As compared with significant leaf growth,inhibition root growth has been detected in 20% natural additive of tomato.Moreover,MS solution with 2%banana has been applied to promote stem growth,by control with notable root proliferation in MS solution with 1%coconut juice.Interestingly,rapid potential growth has been suffered from MS solution with 15%potato and 1% coconut juice in B.striata.After sows the B.striataseed on the medium of MS+2.0mg/L IBA+30g/L sucrose+7g/L agar+15% potato for germination with 30 days,the plantlet is subcultured on the medium of MS+2.0mg/L IBA+30g/L sucrose+7g/L agar+1%coconut juice for 3months,the final cultured seedlings could totally favor for the field culturing with stout stem,sturdy roots and vigorous growing.
