正交实验及响应面法在银杏愈伤组织黄酮提取条件优化中的比较

【摘要】 以银杏愈伤组织为材料,运用正交实验和响应面法对银杏黄酮的提取条件进行优化,并对二者结果进行比较。正交实验的最佳条件是:提取剂选用50%乙醇,微波辐射60 s,70℃水浴浸提2 h,此条件下的黄酮得率为(24.18±0.20)mg/g。响应面实验结果显示,提取剂为46.39%乙醇,微波辐射62.23 s,70℃水浴浸提2 h,黄酮得率达到(24.55±0.28)mg/g,与预测值24.57 mg/g相符。双样本T检验结果表明,响应面最优条件下的黄酮得率较正交实验高1.7%,但差异不显著。

【Abstract】 Using ginkgo callus powder as material,orthogonal test and response surface analysis were carried out to optimize the extracting conditions of ginkgo flavonoids. And the comparison of the results between orthogonal test and response surface test was done. The optimal conditions based on orthogonal experiment were: extraction solvent 50% ethanol,microwave processing 60 s,water bath processing 2 h at70 ℃. Under such conditions,the extraction rate of flavonoids was( 24. 18 ± 0. 20) mg / g. The results of response surface analysis showed that: extraction solvent 46. 39% ethanol,microwave processing 62. 23 s,water bath processing 2 h at 70 ℃,the extraction rate of flavonoids reached( 24. 55 ± 0. 28) mg / g,which matched the predicted value 24. 57 mg / g very well. The results of two-sample T test showed that the extraction rate under the optimal conditions of response surface analysis was 1. 7% higher than that under the optimal conditions of orthogonal test,but the difference was not significant.
