【摘要】 为研究庄河大骨鸡抗粘液病毒基因(myxovirus resistant,Mx)和溶菌酶基因(1ysozyme,LYZ)的多态性,本试验从大骨鸡血液中提取DNA,PCR扩增Mx和LYZ基因后分别进行RsaⅠ和HindⅢ酶切及琼脂糖凝胶电泳。研究结果表明,所选取的262只庄河大骨鸡Mx基因片段均能得到PCR扩增产物,产物大小为101 bp,经酶切后获得GG、GA和AA 3种基因型,基因型频率分别为0.744、0.229、0.027,因此大骨鸡Mx基因具有多态性;所选取的120只大骨鸡LYZ基因片段也均能得到PCR扩增产物,产物大小为590 bp,经酶切后仅获得一种基因型GG型,因此该基因此酶切位点上无多态性。本研究为庄河大骨鸡资源保存、品种选育和开发利用提供了科学依据。更多还原

【Abstract】 To analyze the polymorphism of myxovirus resistant(Mx)and lysozyme(LYZ)gene for Zhuanghe Dagu chicken,The technology of PCR-RFLP was used.The PCR products were digested with enzyme RsaⅠ and Hind Ⅲ.Hind Ⅲ locus polymorphism of LYZ in Zhuanghe Dagu chicken was not detected,RsaⅠ locus polymorphism of Mx was divided into three kinds of genotypes of GG,GA and AA,and the frequencies of genotypes were 0.744、0.229 and 0.027.The research provided the scientific basis for the resource preservation,variety breeding and the development and utilization of Zhuanghe Dagu chicken.更多还原
