Smad-4在结直肠癌的表达及评价患者预后的研究
Clinical outcomese expression of Smad-4 in colorectal cancer tissues,its signal mechanism and its role in the evaluation of patients’ clinical outcomes
【摘要】 目的:借助免疫组化方法检测结直肠癌及癌旁组织中Smad-4的表达情况,评估Smad-4作为结直肠癌患者预后评估指标的可能性。方法:采用免疫组化法检测97例结直肠癌组织及37例癌旁组织中Smad-4的表达情况,结合患者的临床随访资料数据回顾性分析患者肿瘤标本中Smad-4的表达量与患者术后总体无瘤生存两者之间的关系。结果:一定比例的(63例)结直肠癌肿瘤组织中可见Smad-4蛋白的显著表达,其阳性表达率为64.94%[63/97],高于癌旁、正常结直肠黏膜细胞,在结直肠癌旁组织中的阳性表达率为(6例)16.21%[6/37],两者比较,差异具有显著的统计学意义(P<0.05);而且证实Smad-4蛋白表达高、低两组患者的术后生存情况有显著差异(P<0.05)。结论:Smad-4蛋白在结直肠癌患者组织中存在高表达,在患者肿瘤组织中存在Smad-4高表和低表达的两类患者中,其临床预后存在显著的统计学差异。Smad-4蛋白可以有条件的作为评估术后结直肠癌患者临床预后的蛋白标记物。
【Abstract】 Objective: To detect colorectal cancer and paracancerous tissues Bby means of immunohistochemical Smad- 4 method of detecting colorectal cancer and paracancerous tissues,so as to assesssment of the potential of Smad- 4 as the prognosis of patients with colorectal cancer assessment index. Methods: Expression of Smad- 4was detected by immunohistochemistry in 97 cases of colorectal carcinoma tissues and 37 cases of para carcinoma tissues,combined with clinical follow- up data of a retrospective analysis of expression of Smad- 4 in tumors from patients in patients after general no relationship between survival of both. Results: Certain proportion( 63 cases) was significantly expressed Smad- 4 protein in colorectal cancer tissues,the positive expression rate of [63/97] was 64. 94%,higher than that of adjacent normal colorectal mucosa,the positive expression in cells,tissues adjacent to colorectal cancer in the rate of( 6 cases) 16. 21% [6/37],( P < 0. 05). The comparison between the two was,different with statistical significance significant differences in the expression of Smad- 4 protein and confirmed that; high,low of the two groups of patients with postoperative survival have significant differences( P < 0. 05). Conclusions: Smad- 4 protein has high expression in tissue of patients with colorectal cancer,there are two kinds of patients with higher Smad- 4 surface and low expression in cancer tissue,there was a significant difference,the clinical prognosis. Protein marker Smad- 4 protein can be used to evaluate the clinical outcome of patients with rectal cancer after operation nodes of conditional.
