2型糖尿病大鼠破骨细胞变化

【摘要】 目的观察2型糖尿病大鼠破骨细胞的变化。方法 40只2.5~3个月龄雌性Wistar大鼠随机分为2型糖尿病模型组和正常对照组各20只。2型糖尿病成模后第0、4、8、12周末各时点选取5只大鼠测空腹血糖(fasting blood glucose,FBG)、空腹胰岛素(fasting serum lisulin,FINS);第0、4、8、12周末各组随机选取5只大鼠并收集骨髓细胞行体外破骨细胞培养、破骨细胞染色和计数。结果与正常组相比,糖尿病组大鼠血糖及胰岛素水平显著性升高,破骨细胞计数显著性增高。糖尿病组大鼠破骨细胞数量与胰岛素敏感指数呈负相关。结论 2型糖尿病大鼠破骨细胞计数增多可能是其骨量降低的重要原因。

【Abstract】 Objective To observe the change of osteoclasts in type 2 diabetes mellitus(T2DM)rats.Methods Forty 2.5-3 months female Wistar rats were randomly divided into T2DM group and normal control group.Blood glucose and insulin were measured in T2DM model rats copied successfully at 0,4,8,12 th weekend;and at 0,4,8,12 th weekend,the osteoclasts of five rats randomly selected from each group were collected for in vitro osteoclast culture,osteoclasts staining and counting.Results Compared with the normal group,the blood glucose and insulin levels of T2DM group significantly increased,the osteoclasts count increased significantly.The osteoclasts number of T2DM group was negatively correlated with insulin activation index.Conclusion The increasing osteoclast count may be one of the important reasons for bone loss in T2DM rats.
