阿奇霉素联合左氧氟沙星对大肠杆菌生物膜的干预

【摘要】 目的:研究阿奇霉素(AZM)联合左氧氟沙星(LFX)对腹膜透析(PD)管大肠杆菌(E.coli)生物膜(BF)的影响。方法:(1)二倍稀释法分别测定AZM、LFX对临床分离E.coli最低抑菌浓度(MIC)及最低杀菌浓度(MBC);(2)建模3d后对各组分别进行处理。A组:空白对照组;B组:1/8AZM MIC组;C组:1LFX MBC组;D组:1/8AZM MIC+1LFX MBC组。经药物作用12h,各组分别以连续稀释法行BF内活菌计数,扫描电子显微镜观察PD管表面BF结构。结果:(1)B组与A组比较差异无统计学意义(P>0.05);C组与A组比较活菌数量减少,差异有统计学意义(P<0.01);D组与C组比较活菌数量减少,差异有统计学意义(P<0.05);(2)扫描电子显微镜观察可见D组E.coli BF结构体积较A组减少。结论:AZM可增强LFX对PD管表面E.coli BF内细菌抗菌效应。

【Abstract】 Objective:To investigate the destructive effects of Azithromycin(AZM)and Levofloxacin(LFX)to E.coli biofilm of the dialysis catheter tube,and its bactericidal effect of BF within E.coli.Methods:(1)Detected the Minimum inhibitory concentration(MIC)and Minimum bactericidal concentration(MBC)of AZM and LFX on E.coli.(2)Divided into four groups—Group A(lank control group),Group B(1/8MIC AZM group),Group C(1 MBC LFX group),Group D(1/8 MIC AZM and 1 MBC LFX group).After 12-hour medication,SEM observed the surface of the dialysis catheter tube’s BF,and crystal violet staining BF semi-quantitative,using flat-panel micro-quantitative method to count viable cells on the surface of the carrier.Results:(1)After 12-hour medication,we used flat-panel micro-quantitative method to count cells on the surface and found that there was no statistically significant between Group A and Group B(P >0.05);the bacteria reduced in Group C compared with Group A(P <0.01);the bacteria were reduced in Group D compared with Group C(P <0.05).(2)We used scanning electron microscopy to observe surface E.coli BF structure and size,and found that the bacteria reduced significantly in Group D than in Group A(P <0.05).Carrier of silver staining shows cotton-like BF.Conclusion:The killing effect of combination of AZM and LFX was significantly reduced.
