抗菌肽-人溶菌酶融合基因的构建及转入锦橙后的表达分析
Construction and Expression Analysis of Cecropin B and Human Lysozyme Fusion Gene in Transgenic Citrus
【摘要】 为了研究抗菌肽和人溶菌酶对柑桔溃疡病和黄龙病等的协同抗性,利用连接多肽2A编码序列连接抗菌肽cecropin B(CB)和人溶菌酶human lysozyme(HLy)基因,构建了融合基因CB:HLy及其超量表达载体p35S::CB:HLy。利用农杆菌介导法,将该载体转化至锦橙上胚轴。通过GUS化学染色及PCR鉴定,获得转基因植株32株。Real-time PCR证明了融合基因在转基因锦橙中得到了高效表达。本研究为柑桔溃疡病的抗性研究提供了材料。
【Abstract】 To investigate the synergy resistance of antibacterial peptide and human lysozyme to citrus canker and HLB,we constructed a CB:HLyfusion gene and its overexpression vector p35S::CB:Hly using one linker polypeptide 2A.This vector was introduced into sweet orange genome(Citrus sinensis Osbeck)by Agrobacterium-mediated transformation.Thirty-two transgenic plants were confirmed by GUS chemical staining and PCR analysis.Real-time PCR showed that this fusion gene had high expression levels in the transgenic citrus.This study provided the materials for research on resistance to citrus canker in the future.
