双重PCR鉴别牛、羊肉掺假
Identification of adulteration in mutton and cattle by duplex PCR
【摘要】 目的建立可同时检测羊肉中掺入的猪、马、牛、鸭成分中的任意2种,及牛肉中掺入的猪、马、鸭成分中任意2种的双重PCR检测体系。方法配制掺入猪、马、牛、鸭源性成分的模拟掺假羊肉样品及掺入猪、马、鸭源性成分的模拟掺假牛肉样品,每个成分的掺假比例为10%。筛选特异性强、敏感度高的猪、马、牛、鸭物种特异性基因,优化反应体系,建立了针对猪、牛、马、鸭两两配对的双重PCR检测体系。结果无论是模拟牛肉掺假样品,还是模拟羊肉掺假样品,所有掺入的低价肉品成分都被准确检出,且无非特异扩增条带。结论本研究建立的双重PCR检测体系,可以敏感、特异、准确的检出牛羊肉制品中的猪、牛、马、鸭成分。
【Abstract】 Objective To establish duplex PCR system to simultaneously detect pig,horse,cattle and duck mixed in mutton or detect pig,horse and duck mixed in cattle. Methods Mutton and cattle samples were mixed with 10% pig,horse,duck or cattle( only in mutton samples). The species genes specific for pig,horse,cattle and duck were selected for good specificity and sensitivity to form duplex PCR system. Results The duplex PCR system can identify single added component whatever it is component in mutton or in cattle accurately and there is not nonspecific bands. Conclusion The established duplex PCR system can detect pig,horse,cattle and duck mixed in mutton and cattle sensitively,specifically and accurately.
