O/T百合脱毒技术研究

【摘要】 以感染郁金香碎锦病毒(TBV)和百合无症病毒(LSV)的O/T系百合黄天霸(Manissa)为试验材料,进行了茎尖诱导培养基的筛选;采用不同大小茎尖培养和热处理结合茎尖培养的方式脱除病毒;运用RT-PCR技术进行病毒检测。结果表明,MS+6-BA0.5mg/L+NAA0.3mg/L培养基比较适宜茎尖不定芽的诱导;(39±1)℃热处理结合0.5~0.7 mm大小的茎尖培养脱毒效果最好。

【Abstract】 Bulbs of O/T style lily Manissa which were infected by TBV and LSV were used as material in this experiment.Media selection of adventitious buds induction from stem tip was carried on by methods of stem tip culture and heat treatment combined with stem tip culture,and the viruses were detected by RT-PCR. The results indicated that optimum media for stem tip induction was MS+6-BA 0.5 mg/L +NAA 0.3 mg/L. The virus-elimination can get best efficiency at the condition that the plantlets were forcing heated by(39±1) ℃, combined with stem tip culture which were 0.5~0.7 mm.
