游泳运动和白藜芦醇对Ⅱ型糖尿病大鼠肝糖原及肝脏GLUT2的影响

【摘要】 目的:研究游泳运动和白藜芦醇(Res)对Ⅱ型糖尿病(T2DM)大鼠糖代谢及肝脏CLUT2的影响。方法:采用5周高脂饮食加小剂量链脲佐菌素(35 mg/kg)构建T2DM大鼠模型。32只健康SPF级雄性SD大鼠随机分成正常组(C组)、正常运动组(CE组)、正常Res组(CR组)和正常运动+Res组(CER组),32只T2DM大鼠随机分为模型组(D组)、T2DM+运动组(DE组)、T2DM+Res组(DR组)和T2DM+运动+Res组(DER组),每组均8只。采用游泳运动训练,每周6天;Res灌胃处理每周7天。连续干预7周后,检测各组大鼠空腹血糖(FBG)、口服葡萄糖耐量试验(OGTT)、肝糖原;Western blot检测肝脏GLUT2蛋白表达。结果:T2DM大鼠FBG以及OGTT后各阶段血糖浓度较C组均非常显著升高(P<0.01),DE组、DR组、DER组OGTT后30 min、60 min及120 min血糖较D组均非常显著下降(P<0.01),DER组与DE组、DR组比较进一步降低(P<0.01);与C组比较,CE组和CR组大鼠肝脏GLUT2表达显著升高(P<0.05或P<0.01),CER组大鼠肝糖原、GLUT2蛋白表达非常显著升高(P<0.01),D组肝糖原、GLUT2蛋白非常显著下降(P<0.01)。与D组比较,DE组、DR组和DER组肝糖原、GLUT2蛋白表达均非常显著降低(P<0.01),其中DER组进一步降低(P<0.05)。结论:运动训练和白藜芦醇能降低T2DM大鼠糖耐量受损程度,增加肝糖原储备,改善葡萄糖代谢,两者联合干预较单一效果更显著,其机制可能与肝脏GLUT2蛋白表达升高有关。

【Abstract】 Objective To explore the effects of aerobic exercise and administration of resveratrol(Res)on the carbohydrate metabolism and the expression of GLUT2 in the liver of rat with type 2 diabetes mellitus(T2DM). Methods 32 rats were fed with high-carbohydrate and high-fat diet and given intraperitoneal injection of streptozotocin(STZ)with the dosage of 35mg/kg for 5 weeks to induce T2DM, then they were randomly divided into following groups:T2DM(D),T2DM exercise(DE),T2DM Res administration(DR),T2DM exercise plus Res(DER). Another 32 healthy SD rats were randomly divided into 4 groups:control(C),exercise(CE),Res administration(CR),and exercise plus Res administration(CER). Rats in all exercise groups underwent swimming 6 times per week for 7 weeks. Rats in Res administration groups were orally administrated Res with the dosage of 45mg/kg per day for 7 weeks. Fasting blood glucose(FBG),oral glucose tolerance test(OGTT),hepatic glycogen,and the expression of GLUT2 in liver were determined after 7-week experiment. Results The FBG and concentration of blood glucose at 30,60,and 120 minutes after OGTT in group D were higher than that in group C(P < 0.01), and those in groups DE,DR,and DER were lower than the group D(P < 0.01). The FBG and the concentration of blood glucose at 30,60,and 120 minutes after OGTT in group DER were significantly lower than that in groups DE and DR(P < 0.01). Comparing with group C,the expression of GLUT2 in the liver of groups CE and CR increased significantly(P < 0.05 or P < 0.01),and the expression of GLUT2 and the content of hepatic glycogen in group CER increased significantly(P < 0.01),and those in group D decreased significantly(P < 0.01). Comparing with group D,the expression of GLUT2 and the content of hepatic glycogen in groups DE,DR,and DER decreased significantly(P < 0.01),especially in group DER. Conclusions Exercise combined with administration of resveratrol improves the impaired glucose tolerance of T2DM rats,increases the content of hepatic glycogen,and ameliorates the carbohydrate metabolism more significantly than exercise or administration of resveratrol alone.
