人参皂苷Rb1对缺血再灌注大鼠肝损伤的保护作用
Protective effects of ginsenoside Rb1 for liver ischemia-reperfusion injury in rats
【摘要】 [目的]探讨人参皂苷Rb1(GS-Rb1)对缺血再灌注大鼠肝损伤的保护作用及其量效关系.[方法]采用Pringle法建立全肝入肝血流阻断肝缺血再灌注损伤模型,GS-Rb1小、大剂量组大鼠在阻断前经肠系膜上静脉注射给予不同剂量(20,40mg/kg)GS-Rb1溶液.肝脏缺血20min后松开血管夹进行再灌注,于再灌注2h后采集下腔静脉血,并取肝组织标本.检测血清中ALT,AST水平,WST-1法测定血清中SOD活性,TBA法测定血清中MDA含量.肝组织标本行HE染色后置于光学显微镜下观察病理形态学变化.[结果]与正常对照组比较,GS-Rb1小剂量组血清中ALT,AST水平均明显下降(P<0.05),血清中SOD活性明显升高(P<0.05),MDA含量明显降低(P<0.05);人参皂苷Rb1小、大剂量组肝组织病理损伤减轻.[结论]合适剂量的GS-Rb1(20mg/kg)可对缺血再灌注肝损伤起到保护作用,而保护作用不随剂量的增加而增强.
【Abstract】 OBJECTIVETo study the protective effects and dose-effect relationship of ginsenoside Rb1 for liver ischemia-reperfusion injury in rats.METHODS Model of liver ischemia-reperfusion injury in rats were established by Pringle’s method.20,40mg/kg of ginsenoside Rb1 solution in small and large dose groups were injected to the superior mesenteric vein of rats before blocking.The vascular clamp was loosen to proceed the reperfusion after liver ischemia for 20 min,and the venous blood from inferior vena cava and liver tissues were taken out after reperfusion for 2h.The alanine aminotransferase(ALT)and aspartate aminotransferase(AST)were measured in serum,and the content of malondialdehye(MDA)was detected by TBA,and activity of superoxide dismutase(SOD)were measured by WST-1.The histopathologic changes of liver tissue were observed.RESULTS Compared with normal control group,the contents of ALT,AST and MDA decreased significantly(P<0.05),and the activity of SOD increased significantly(P<0.05)in small dose group,and the pathological lesions of liver tissue were attenuated in small and large dose groups.CONCLUSION Ginsenoside Rb1(20 mg/kg)could protect liver ischemia-reperfusion injury,and this protective effects have no dose-effect relationship.
