Balb/c小鼠滴鼻感染鼠疫菌的敏感性测定

【摘要】 目的以滴鼻感染的方式对Balb/c小鼠进行鼠疫强毒株敏感性实验。方法将鼠疫菌141标准株37℃24 h培养物,用灭菌生理盐水研磨制成均匀菌悬液,标准比浊后稀释成不同剂量组,分别滴于Balb/c小鼠鼻孔处,攻击感染后连续观察22 d,记录小鼠特异性死亡。同时进行平板计数,以菌落形成单位可推算出实际攻击活菌数量。结果活菌计数结果:培养48 h后7个平板菌落形成单位平均为23.6 cfu,强毒鼠疫菌141株滴鼻感染Balb/c小鼠的绝对致死量为118 000 cfu。结论 Balb/c小鼠经滴鼻感染后对鼠疫菌的敏感性远远低于皮下感染,故鼠疫菌滴鼻感染Balb/c小鼠的方式在鼠疫鉴定和鼠疫疫苗方面的研究中具有重要意义。

【Abstract】 Objective To test the sensitivity of Balb/c mice which were infected by intranasal routes. Methods The 141 standard plague strains were cultured in 37℃, 24h and grinded into bacteria levitation liquid. Used the turbidimetric method diluting the bacteria levitation liquid into different dosage groups, dripped the liquid to the nostrils of Balb/c mice, respectively. Challenged the Balb/c mice with 141 strain and continuously observed for 22 days, recorded the specific death results. Meanwhile, counted the bacteria on the plate and we could predict the actual infected number of viable count. Results After 48h, the average CFU in 7 culture vessels was 23.6, absolute lethal dose of Balb/c mice was 118 000 viable count. Conclusion The sensitivity of Balb/c mice which are infected with Yersinia pestis by intranasal routes is far below intramuscular routes, so intranasal infection method has an important implication for plague identification and vaccine research.
