精液中纳米细菌检测方法的建立
Study on nanobateria detection method applied for semen examination
【摘要】 目的建立简便、快速、高效检测精液中纳米细菌的荧光定量RT-PCR法。方法采取12例睾丸微石症患者精液标本及12例健康人正常对照精液标本,进行PCR检测,并对RT-PCR重复性,灵敏度,特异性验证。结果 12例睾丸微石症患者精液标本中,6例标本出现阳性,阳性率占50%,健康正常对照精液标本检出1例阳性。建立RT-PCR一步法检测精液中纳米细菌,灵敏度达到500 PEU/ml,Ct变异系数均<5%,重复性好。结论荧光定量RT-PCR法检测睾丸微石症患者精液纳米细菌方法可靠,简便准确,稳定性好。
【Abstract】 Objective To establish a simple,fast and efficient detection method with nanobacteria fluorescent quantitative RT- PCR assay. Methods The semen samples of 12 patients with testicular microlithiasis and semen specimens of 12 healthy controls were examined by PCR assay,and RT- PCR assay had been established with characteristics of authenticity,repeatability and sensitivity. Results In semen specimens of 12 patients with testicular microlithiasis,6 specimens were positive,and the positive rate was 50%. In healthy and normal controls,only 1 specimen appeared positive,the sensitivity of RT- PCR one- step detection of nanobacteria reached 50 PEU / ml. CV of RT- PCR one- step for detection of nanobacteria was < 5%. Conclusion The effectiveness of fluorescent quantitative one- step RT- PCR in detection of nanobacteria is reliable,simple,accurate and stable.
