三磷酸腺苷与骨髓间充质干细胞联合移植对损伤脊髓中神经元特异性烯醇化酶表达的影响
Effect of neuron specific enolase on injured spinal cord by transplanting adenosine triphosphate and bone marrow mesenchymal stem cells
【摘要】 目的探讨三磷酸腺苷(ATP)与骨髓间充质干细胞(BMSCs)复合物联合移植对损伤脊髓的修复及脊髓中神经元特异性烯醇化酶(NSE)表达的影响。方法将48只SD雌性大鼠采用改良Allen’s打击法制备T12节段脊髓损伤模型后,随机分为4组,12只/组,A组(ATP+BMSCs联合移植组)、B组(BMSCs移植组)、C组(ATP移植组)、D组(空白对照组)。术后采用Rivlin斜板实验评价后肢功能恢复程度,于28 d处死大鼠后取材行组织学和免疫组织化学染色,观察NSE的表达和脊髓损伤修复情况。结果术后各组动物均出现后肢瘫痪症状,术后23周开始有不同程度恢复,A组恢复效果最佳,B、C组次之,D组最差。Rivlin斜板实验评分:术后14、21及28 d,A组NSE阳性细胞面积百分比高于B、C、D组,B、C组高于D组,差异均有统计学意义(P<0.05);B、C组间比较差异无统计学意义。结论 ATP与BMSCs复合物联合移植使损伤脊髓中神经元NSE表达增加,且肢体功能恢复效果最佳,有利于脊髓损伤的修复。
【Abstract】 Objective To study the effect of acute injured spinal cord repair and the expression of neuron specific enolase(NSE) by transplanting adenosine triphosphate(ATP) and bone marrow mesenchymal stem cells(BMSCs) in injured spinal cord(SCI).Methods 48 experimental adult female SD rats(weighing 240-260 g) were made SCI models at T12 levels according to the improved Allen’s method and were divided into 4 groups randomly.Group A,with ATP(40 mg/kg) and BMSCs(6 μL) injection to the SCI;group B,with only BMSCs injection;group C,with only ATP injection;group D,only laminectomy.Their general conditions were observed.The nerve function of low extremities was evaluated using the Rivlin inclined plane test at 14,21 and 28 days after transplantation.After 4 weeks,rats were sacrificed,and their spinal cords were examined for NSE by immunohistochemical staining.Results Paralysis symptom in hind limb occurred after operation in all experimental rats and began to recover since 2 to 3 weeks postoperatively.Group A was the best,group B and C were better,and group D was the worst.The Rivlin inclined plane test showed that 14,21 and 28 days after operation,the comparison between group A,B,C and D,except between group B and C,had significant statistical differences(P <0.05).The ultrastructures of spinal cord tissue showed that there were immunohistochemical positive staining cells with antibody of NSE in groups A,B and C at 28 days after operation,and positive cells of group A was significantly more than that of group B and C(P <0.05).There weren’t positive staining cells in group D.Conclusion ATP and BMSCs can increase the expression of NSE by transplanting it to SCI and can be beneficial to the recovery of SCI.
