人类多重剪接RNA结合蛋白类基因RBPMS2的表达分析
Expression pattern and subcellular localization of human RBPMS2
【摘要】 应用RT-PCR扩增技术建立人类多重剪接RNA结合蛋白类基因RBPMS2在人体内(肝、脑、心脏、肺、胃、肾、胎盘、膀胱、骨骼肌、结肠、睾丸、前列腺、卵巢、子宫)的组织表达谱,组织表达谱显示RBPMS2基因在上述组织中均有表达,且在心脏、骨骼肌组织中表达尤为显著。为进一步研究该基因蛋白的表达情况,通过构建pEGFP-C1/RBPMS2融合表达载体转染细胞进行亚细胞定位,结果表明:RBPMS2在细胞的核质中均有分布,且在细胞核中分布相对较少。对RBPMS2基因进行的组织表达谱及亚细胞定位,为进一步研究RBPMS2的生物学功能奠定了基础。
【Abstract】 The study was to establish the tissue expression profile of RBPMS2(NM 194272) gene in human tissue(lung,placenta,prostate,ovary,kidney,liver,heart,bladder,skeleton muscle,brain,colon,testis,uterus,stomach)by the technology of RT-PCR.The results showed that RBPMSZ gene expressed in these tissues and the expression was remarkable in heart and skeleton muscle.To further study the expression of RBPMS2 protein,we instructed a fusion expression vector of pEGFP-Cl/RBPMS2 and transfected it into COS7 cell.The subcellular localization showed that the distribution of RBPMS2 was in the nucleus and cytoplasm,but with a minority distribution in the nucleus.The establishment of tissue expression profile and subcellular localization of RBPMSZ lay a good foundation for the futher sdudy of RBPMS2 biological function.
