依替米贝中间体(4S)-3-[5-(4-氟苯基)-1,5-二氧代戊基]-4-苯基-2-噁唑烷酮的合成
Synthesis of(S)-1-(4-Fluorophenyl)-5-(2-oxo-4-phenyloxazolidin-3-yl)pentane-1,5-dione-An Intermediate for Ezetimibe
【摘要】 以氟苯、戊二酸酐等为原料,经历付克反应得到5-(4-氟苯基)-5-氧代戊酸,再与S-辅基等缩合得到目标化合物(4S)-3-[5-(4-氟苯基)-1,5-二氧代戊基]-4-苯基-2-噁唑烷酮。目标产物收率78.2%,高于现有报道69.1%。
【Abstract】 The titled compound is a key intermediate for Ezetimibe. It was synthesized in 78.2% yield starting from fluorobenzene and dihydro-3H-pyran-2,6-dione via intermediate of 5-(4-fluorophenyl)-5-oxopentanoic acid. The reaction conditions were optimized.
