健脾化湿颗粒对IBS-D模型大鼠血清P物质及ET表达的影响
Effect of Jianpihuashi Granules on expression of serum substance P and ET of diarrhea-predominant irritable bowel syndrome rats
【摘要】 目的观察健脾化湿颗粒对腹泻型肠易激综合征(IBS-D)大鼠血清P物质(SP)及内皮素(ET)表达的影响,探讨其治疗机制。方法取清洁级Wistar大鼠60只,平均分为对照组、模型组及健脾化湿颗粒低、中、高剂量组和阳性对照的匹维溴铵组。除对照组外各组大鼠束缚应激配合番泻叶煎剂溶液灌胃复制模型。造模成功后,予以相应药物治疗。实验过程中观察各组大鼠大便性状及大便次数。实验结束后酶联免疫吸附测定(ELISA)观察血清中ET、SP水平变化。结果与模型组比较,各治疗组大鼠腹泻症状明显改善;健脾化湿高剂量组与匹维溴铵组大鼠症状改善更为明显,组间比较差异无统计学意义(P>0.05)。高剂量组、中剂量组、匹维溴铵组与模型组比较,血清SP水平下降明显,差异有统计学意义(P<0.05);高、中剂量组与匹维溴铵组比较差异无统计学意义(P>0.05)。高剂量组、中剂量组、匹维溴铵组与模型组比较,血清ET水平升高明显,差异有统计学意义(P<0.05),其中健脾化湿中、高剂量组组间比较,差异有统计学意义(P<0.05),表明有一定剂量依赖关系。结论健脾化湿颗粒可明显改善IBS-D模型大鼠胃肠功能,其作用机制可能与降低血清SP水平,提高血清ET水平有关。
【Abstract】 Objective To observe the effect of Jianpihuashi(JPHS)Granules on the expression of substance P(SP)and endothelin(ET)in diarrhea-predominant irritable bowel syndrome(IBS-D)rats and to explore its therapeutic mechanism.Methods Sixty clean-level Wistar rats were randomly divided into the control group,model group,low-dose,middle-dose and high-dose JPHS Granules groups and the positive control group(Dicetel group).Except the control group,the other groups were performed the restraint stress combined with folium sennae decoction by gavage to establish D-IBS rat model.After the success of the model construction, the corresponding medication was given.The stool property and times were observed during the experiment.After the end of the experiment,ELISA was applied to detect the change of ET and SP levels.Results Compared with the model group,the diarrhea symptoms in the various medication groups were obviously improved;which in the high-dose JPHS Granules group and the Dicetel group were improved more significantly,but the difference between these two groups had no statistical significance.Compared with the model group,the serum SP level in the high-dose,middle-dose JPHS granules groups and the Dicetel group was obviously decreased,the difference showed the statistical significance(P<0.05).Compared with the model group,serum ET level in the highdose,middle-dose JPHS Granules groups and the Dicetel group was obviously increased,the difference showing statistical significance(P<0.05),in which,the difference between the high-dose and the middle-dose JPHS Granules groups had statistical significance(P<0.05),which indicating that the certain dose-dependence relation existed between them.Conclusion JPHS Granules can obviously improve the gastrointestinal function of IBS-D model rats,its effect mechanism may be related with decreasing the serum SP level and increasing the serum ET level.
