铜藻多糖水提法工艺优化及其抗氧化活性研究

【摘要】 [目的]研究铜藻多糖的水提法提取工艺,并研究其抗氧化活性。[方法]采用水提法提取铜藻多糖,研究提取时间、提取温度和料液比等因素对多糖提取率的影响,以多糖得率为指标,通过正交试验优化最佳提取工艺;并对铜藻多糖的1,1-二苯基苦基苯肼(DPPH)自由基、超氧阴离子自由基(O-2)的清除能力及抗脂质过氧化能力进行研究。[结果]该多糖水提法提取的最优条件为料液比1∶20g/ml、时间3 h、温度60℃,在此条件下,铜藻多糖的提取率为4.9%;铜藻多糖抗氧化活性随着浓度增加而增强,其超氧阴离子(O-2)和DPPH自由基清除能力、抗脂质过氧化能力分别为48.1%、38.8%、34.2%。[结论]该研究优化了铜藻多糖的提取工艺,提取的活性多糖具有一定抗氧化活性,试验优化的工艺稳定可行,为铜藻多糖的后续研究和开发提供依据。

【Abstract】 Extraction technology of polysaccharide from Sargassum herneri(Turn) C. Ag. and its antioxidative activity were studied. [Method]The extraction technology was optimized by orthogonal experiment with the content of polysaccharide which measured by phenol-sulfuric acid method and with the ratio of raw materials to solvent,extracting temperature,extracting time as factors,moreover,1,1-diphenyl-2-picrylhydrazyl(DPPH) free radical,scavenging capacity of superoxide anion radical,anti-lipid peroxidation and reducing power of Sargassum herneri polysaccharide were determined. [Result]The optimal extraction technology of polysaccharide was as follows: the extraction time was 3 h,the extraction temperature was 60 ℃,the ratio of raw material to solvent was 1∶ 20 g /ml. The antioxidative activity Sargassum herneri polysaccharide was increased in a concentration-dependent manner,which the scavenging capacity of the DPPH free radical was38. 8%,scavenging capacity of superoxide anion radical was 34. 2% and the capacity of anti-lipid peroxidation was 48. 1%,respectively.[Conclusion]The optimal extraction parameters was gained in this study and antioxidative activity of Sargassum herneri polysaccharide were investigated,which provided support for the development of Sargassum herneri polysaccharide.
