【摘要】 [目的]采用分子生物学技术,探讨贵州凤岗富锌富硒绿茶对小鼠抗氧化、抗衰老的作用及机制。[方法]腹腔注射D-半乳糖建立小鼠衰老模型,空白组给予腹腔注射0.9%生理盐水。造模14 d后,根据中国营养学会推荐每日饮茶量及实验动物药物剂量比例,给小鼠灌胃富锌富硒绿茶,1次/d。连续灌胃21 d后,采用Morris水迷宫进行行为学实验。处死小鼠,采用蛋白免疫印迹法(western-blotting)检测各小鼠肝脏组织中超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)的表达。采取RT-PCR法,测定各小鼠肝脏组织中SOD和GSH-Px的mRNA转录水平。[结果]富锌富硒绿茶对衰老小鼠学习记忆能力有一定的改善作用,并且提高SOD和GSH-Px的mRNA及蛋白质表达水平(P<0.01)。[结论]富锌富硒绿茶提高机体的抗氧化能力及免疫功能,在一定程度上具有延缓衰老的作用。更多还原

【Abstract】 [Objective]This paper focused on discussing how the Se-enriched and Zinc-enriched green tea works on mice in anti-oxidation and anti-aging by measuring the mRNA and protein expression level of SOD and GSH-Px in liver tissue. [Method]The mice aging model was constructed by intraperitoneal injection of D-galactose,compared with intraperitoneal injection for control group. 14 days later,the experimental mice were fed with tea solution by intragastric administration according to the daily amount of drinking tea and drug dose ratio of experimental animals recommended by Chinese Nutrition Institute. After 3-week gavage,the behavior test was conducted through Morris’ water maze method. The mRNA expressional level of SOD and GSH-Px inside the liver tissue were examined by RT-PCR. The protein expression level of SOD and GSH-Px inside the liver tissue were examined by western-blotting. [Result]The Se & Zinc-enriched green tea can exert a good influence on the improvement of the aging mice’s learning and memorizing ability. Meanwhile,Se & Zinc-enriched green tea can increase mRNA and protein expression levels of SOD and GSH-Px. [Conclusion]Drinking Se & Zinc-enriched can improve the antioxidant capacity of mice and to an extent can slow down the aging process.更多还原
