脉络宁针剂致敏RBL-2H3细胞的实验研究
Study of Mailuoning Cause an Allergic the RBL-2H3 Cells
【摘要】 目的:通过研究临床含有绿原酸的中药注射剂脉络宁针剂对RBL-2H3细胞(大鼠嗜碱性白血病细胞系)脱颗粒、释放介质的影响,探讨绿原酸与中药注射剂发生严重过敏反应的关系,为临床安全用药提供依据。方法:以含有绿原酸的中药针剂脉络宁为研究对象,以不含绿原酸的葛根素针剂为阴性对照,以绿原酸对照品为阳性对照,通过对体外培养RBL-2H3细胞模型的作用,观察该细胞模型发生脱颗粒反应和细胞形态变化,检测β-己糖胺酶释放量。结果:脉络宁、绿原酸组可显著引起RBL-2H3细胞释放β-己糖胺酶(P<0.05);葛根素注射液组能不同程度促进β-己糖胺酶的释放。结论:脉络宁组可显著引起RBL-2H3细胞释放β-己糖胺酶,提示绿原酸与中药注射剂脉络宁发生严重过敏反应有密切关系。
【Abstract】 Objective:Relativity between the severe allergic reaction of the chlorogenic acid and traditional chinese medicine injection was studied by observing influnce of Mailuoning injection on release media of RBL-2H3 cell(rat basophilic leukemia cell line).Method: Mailuoning containing chlorogenic acid was as a suject,and puerarin injection not containing as a negative control,while chlorogenic acid standard as a positive control,the influntion of RBL-2H3 cells model in vitro,the degranulation response and changes in cell morphology was observed,β-hexosaminidase release was detected.Result: The Mailuoning and chlorogenic acid can be a significant factor resulting in release of β-hexosaminidase from RBL-2H3 cells(n=5,P<0.05),puerarin injection group could promote the release of β-hexosaminidase.Conclusion: The Mailuoning group abversely caused the RBL-2H3 cells releasing β-hexosaminidase,and there is some relativity between the severe allergic reaction of the chlorogenic acid and traditional chinese medicine injection.
