2009~2011年安徽省甲型H1N1流感病毒血凝素基因特征分析

【摘要】 目的分析2009~2011年安徽省甲型H1N1流感病毒血凝素基因HA基因演化特征及分子流行病学特点。方法选定2009~2011年安徽省分离的56株甲型H1N1流感毒株,对HA基因进行测序和比对分析。结果对比A/California/07/2009(H1N1),56株H1N1流感病毒的同源性为98.8%~99.7%,HA1蛋白178、187、193、202、207、220、222位发生氨基酸改变。2011年初流行的甲型H1N1毒株全部带有S202T突变。结论基因进化分析显示安徽省流行的甲型H1N1流感HA高度同源,但某些氨基酸位点发生了突变。HA1的202位氨基酸残基突变可能是引发2011年甲型H1N1流感流行的原因之一。

【Abstract】 Objective To classify and identify one Shigella flexneri strain(GH24) from Qinghai Province at the molecular level.Methods The 16S rRNA gene of GH24 was amplified in vitro and sequenced.Its homology was analyzed using GenBank,MicroSEQ ID,and RDP databases.A phylogenetic tree was then constructed based on multiple sequence alignments of GH24 sequences with other 16S rRNA sequences of Shigella.Results The 16S rRNA gene of GH24 was 1,453 bp in length.The isolates had similarity to Shigella in GenBank(99%) and to S.flexneri in MicroSEQ ID and RDP databases(99.91% and 100%,respectively).Multiple sequence alignments and the phylogenetic tree indicated that GH24 was a strain of S.flexneri.Conclusion The strain GH24 was identified as S.flexneri by 16S rRNA gene sequence analysis,which is rapid and reliable.
