QuilA为佐剂的H1、H3亚型流感病毒多表位DNA疫苗免疫评价

【摘要】 目的选用Quil A为佐剂的H1、H3亚型流感病毒多表位DNA疫苗免疫小鼠,评价免疫效果。方法将pVAX1-H3-EHA-H1HA1new,pVAX1-H3-H1HA new,pVAX1-EHA等3种候选核酸疫苗免疫BALB/c小鼠,进行细胞免疫和体液免疫水平检测。结果体液免疫检测显示,免疫后35d,pVAX1-H3HA-EHA-H1HA1new免疫组小鼠产生H1亚型流感病毒特异性体液免疫应答,且抗体水平略高于其他免疫组。细胞免疫检测显示,当受到H1亚型流感病毒抗原刺激时,各免疫组T淋巴细胞均表现出特异性增殖反应和CD3+CD4+、CD3+CD8+T淋巴细胞增多,各免疫组小鼠血清IL-2、IL-4、IL-10及脾脏淋巴细胞IFN-γ水平显著高于对照组(P<0.05)。结论以Quil A为佐剂的H1、H3亚型流感多表位疫苗可诱导免疫小鼠产生特异性的体液和细胞免疫应答。

【Abstract】 Objective BALB/C mice were used to evaluate the immunogenicity of an H1/H3 combined multi-epitope vaccine with the adjuvant Quil A.Methods BALB/C mice were immunized with the DNA vaccines pVAX1-H3-EHA-H1HA1 new,pVAX1-H3-H1HA new,and pVAX1-EHA.Immunity at the humoral and cellular level was then determined.Results In terms of humoral immunity,mice receiving pVAX1-H3HA-EHA-H1HA1 new had a higher antibody level according to ELISA than did other mice 35 days post-inoculation.In terms of cellular immunity,mice in immunized groups had specific proliferation of T cells and increased CD3+CD4+ and CD3+CD8+ T cells.The sera of immunized mice had higher levels of the cytokines IL-2,IL-4,IL-10 and IFN-γ.Conclusion Results confirmed that combined multi-epitope vaccines had significant advantages in terms of inducing both humoral and cellular immunity.
