人乳头瘤病毒16型E7蛋白检测宫颈癌患者特异血清抗体的初步研究

【摘要】 目的探讨重组人乳头状瘤病毒(HPV16型)E7蛋白在宫颈癌血清学检测中的应用价值,以指导宫颈癌的临床诊断。方法采用原核表达系统制备重组HPV16型E7蛋白,通过间接ELISA方法检测29例宫颈癌患者血清中HPV16E7抗体,58例尖锐湿疣妇女及60名健康妇女的血清标本作为对照。结果宫颈癌组特异性抗体水平显著高于尖锐湿疣及健康对照组,3组抗体均值分别为0.63±0.44和0.27±0.10、0.22±0.13,抗体检出阳性率分别为31.0%、5.2%、5.0%;宫颈癌组中HPV16-DAN阳性患者抗体均值明显高于HPV16-DAN阴性者,两者差异有统计学意义(P<0.05)。结论 HPV16型E7蛋白抗原性较强,可作为宫颈癌的血清学诊断候选抗原,对宫颈癌诊断具有的临床应用价值。

【Abstract】 OBJECTIVE To evaluate the application of recombinant HPV16 E7 protein in serology detection of the invasive cervical cancer patients so as to guide the clinical diagnosis of the cervical cancer.METHODS The recombinant protein was expressed in E.coli and purified,which was applied to detect HPV16 E7 antibodies from the serum of 29 invasive cervical cancer patients through indirect ELISA,meantime,the serum samples from 58 women with condylyoma acuminatum and 60 healthy female were set as the positive controls.RESULTS The expression level of specific antibody in the cancer group was significantly higher than that in the positive control group,the average value of the antibodies of the three groups were 0.63±0.44,0.27±0.10,and 0.22±0.13,respectively,and the detection rates of the antibodies were 31.0%,5.2%,and 5.0%,respectively.In the cervical caner group,the average level of antibody was significantly higher in the HPV16 DNA-positive patients than in the HPV16 DNA-negative patients,the difference was statistically significant(P<0.05).CONCLUSION The result shows that the recombinant protein possesses strong antigenicity and can be applied as a candidate antigen for serologic diagnosis,it is of great significance in the clinical diagnosis of cervical cancer.
