【摘要】 目的研究HIP-55在β-肾上腺素受体激动剂异丙基肾上腺素(Isoproterenol hydrochloride,ISO)诱导心脏成纤维细胞活性氧(reactive oxygen species,ROS)产生中的作用。方法分离培养乳大鼠心脏成纤维细胞,利用HIP-55腺病毒和HIP-55-shRNA腺病毒感染乳大鼠心脏成纤维细胞,获得过表达HIP-55或敲减HIP-55的心脏成纤维细胞。ISO刺激后,采用红色荧光探针dihydroethidium(DHE)以及绿色荧光探针二氯荧光黄双乙酸钠(2’,7’-dichlorofluoresce in diacetate,DCFH-DA)的方法检测细胞ROS的产生量。结果过表达HIP-55明显的抑制心脏成纤维细胞ROS的产生,相反敲减HIP-55明显促进心脏成纤维细胞ROS的产生。结论 HIP-55明显抑制ISO诱导的乳大鼠成纤维细胞的ROS的产生。

【Abstract】 Objective The purpose of this study was to determine the role of HIP-55 in ISO-induced reactive oxygen species(ROS) generation in cardiac fibroblasts. Methods Neonatal rat cardiac fibroblasts were cultured and infected with adenovirus HIP-55 or HIP-55-shRNA adenovirus to overexpress or knock down HIP-55. The ROS generation in these cells was detected with red fluoerscent probe dihydroethidium(DHE) and green fluoerscent probe 2’,7’-dichlorofluorescein diacetate(DCFH-DA) after stimulated with ISO. Results ROS generation significantly increased in HIP-55 overexpressed cardiac fibroblasts and significantly decreased in HIP-55 knockdown cardiac fibroblasts. Conclusion HIP-55 can inhibit ISO-induced ROS generation in cardiac fibroblasts.
