高效液相色谱法测定野鸦椿药材中鞣花酸含量

【摘要】 目的建立测定野鸦椿药材中鞣花的含量高效液相色谱法。方法色谱柱为Waters symmetry C18柱(150 mm×4.6 mm,5μm),流动相为甲醇-0.1%磷酸(36∶64),流速1 mL/min,柱温为40℃,检测波长为254 nm。结果鞣花酸进样量在0.129~2.06μg范围内与峰面积呈良好线性关系(r=0.999 4),平均加样回收率为100.57%,RSD为2.43%(n=6)。结论该方法线性关系、精密度和准确度良好,专属性强,可作为野鸦椿药材中鞣花酸的质量控制方法。

【Abstract】 Objective To establish a HPLC method for the determination of the ellagic acid content in Euscaphis Japonica( Thunb.).Methods The chromatographic separation was performed on the Waters symmetry C18column( 150 mm × 4. 6 mm, 5 μm) with methanol-0. 1% H3PO4solution( 36 ∶ 64) as the mobile phase,the flow rate was 1 mL / min and the column temperature was 40 ℃. The detection wave length was 245 nm. Results The linearity of ellagic acid was in the range of 0. 129-2. 06 μg( r = 0. 999 4),the average recovery rate was 100. 57%, RSD was 2. 43%( n = 6). Conclusion This method has good linearity,precision and accuracy strong specificity. It can be used for the quality control method of ellagic acid in Euscaphis Japonica( Thunb.).
