野生茄转化酶抑制子基因StINH1的克隆与序列分析
Cloning and Sequence Analysis of StINH1 Invertase Inhibitor Gene in Solanum torvum
【摘要】 以1个受黄萎病菌诱导的"托鲁巴姆"下调EST为种子序列,在NCBI进行同源搜索,经人工拼接、RT-PCR克隆与序列分析验证,获得野生茄"托鲁巴姆"转化酶抑制子基因(INH)的全长cDNA序列,命名为StINH1。该基因的开放阅读框全长531bp,编码176个氨基酸,与电子克隆获得的序列完全相同。编码蛋白的分子质量为19.916ku,理论等电点为5.14。序列分析表明,该基因属于植物PMEI超家族,编码的前体蛋白含有1个拥有19个氨基酸的前导肽,同时含有多个磷酸化位点。表达模式分析表明,在黄萎病菌侵染后,StINH1在"托鲁巴姆"根中呈下调表达。
【Abstract】 In this study,a down-regulated EST in Solanumtorvuminduced by Verticillium wilt was used as a querying probe to blast the EST database in GenBank.Based on the assembled homologous cDNA sequences,RT-PCR and sequence analysis,a full-length cDNA sequences of invertase inhibitor gene(INH)was confirmed,designated StINH1.It contains a 531bp open reading frame(ORF)that encodes a 176amino acid protein.The coding protein has a molecular mass of 19.916ku and a theoretical isoelectric point of 5.14.Sequence analysis showed that StINH1 belongs to plant PMEI superfamily,and codes a precursor protein containing a 19aa leader peptide and multiple phosphorylation sites.The expression pattern indicated that StINH1 was down-regulated while Solanumtorvum was infected by Verticillium wilt.
