两亲性壳寡糖衍生物的制备、表征及对疏水性染料TO的包覆

【摘要】 以天然高分子壳聚糖为原料,通过在壳聚糖2位—NH2上引入部分疏水性烷基,在6位—OH和2位—NH2上引入亲水性羟乙基,制得N-月桂基-O,N-羟乙基壳聚糖(LGC)两亲性衍生物,分别用FTIR、1HNMR和元素分析等技术对其结构进行表征.此壳聚糖衍生物在水相中自组装形成胶束,以芘为荧光探针研究了其自聚集行为,结果表明制得的自聚体具有较小的临界胶束浓度(CMC为1.67×10-2,mg/mL),并用此壳聚糖胶束对疏水性染料噻唑橙(TO)进行了包覆,发现包覆后染料的荧光强度显著增强.

【Abstract】 With biopolymer chitosan as raw material,the amphiphilic derivative of N-lauryl-O,N-hydroxyethyl,chitosan(LGC)was synthesized by the introduction of hydrophobic moietie of alkyl groups to —NH2at 2,and the introduction of hydroxyethyl groups as the hydrophilic moieties to —OH at 6 and—NH2at 2. The chemical structure of the derivative was characterized via FTIR,1HNMR and elemental analysis. The LGC could self-aggregate to form micelle with a critical micelle concentration(CMC)at 0.0167mg/mL in the aqueous phase. The hydrophobic dye thiazole orange(TO)was encapsulated with this amphiphilic chitosan derivative. The prepared nanoparticles(LGC/TO)showed a remarkable enhancement of fluorescence intensity compared with TO in aqueous solution.
