运动延缓高脂膳食诱导大鼠胰岛素抵抗形成的机制

【摘要】 目的:观察运动对高脂膳食诱导SD大鼠胰岛素抵抗(IR)形成的干预作用及其相关机制。方法:将SD大鼠分为普食对照组(C组)、普食运动组(E组)、高脂膳食对照组(H组)、高脂膳食运动组(HE组);综合利用FINS、FBG、HOMA-IR、ISI指标评价大鼠运动干预的效果;采用酶联免疫法和实时荧光定量PCR技术观察血清脂联素、内脏脂肪组织脂联素mRNA表达在高脂和运动干预条件下所发生的改变;采用Western blotting方法测定大鼠股四头肌和肝脏AMPKα蛋白表达及其磷酸化水平。结果:1)各组大鼠血清脂联素水平未见显著性差异(P>0.05),H组内脏脂肪组织脂联素mRNA表达显著低于C组(P<0.05);2)各组大鼠股四头肌AMPKα蛋白表达无显著性差异(P>0.05),HE组AMPKα(Thr172)磷酸化水平高于H组43.2%(P<0.05),E组AMPKα(Thr172)磷酸化水平高于C组83.7%(P<0.01);3)各组大鼠肝脏组织AMPKα蛋白表达无显著性差异(P>0.05),HE组AMPKα(Thr172)磷酸化水平高于H组51.1%(P<0.05),其余各组无显著性差异(P>0.05)。结论:内脏脂肪组织脂联素mRNA表达的下调可能参与了高脂大鼠IR的形成。未观察到运动对大鼠血清脂联素水平、内脏脂肪组织脂联素mRNA表达有显著影响,提示运动延缓IR形成与脂联素是否在运动后发生变化的因果关系尚无法定论。运动提高了大鼠骨骼肌和肝脏AMPKα(Thr172)磷酸化水平,提示骨骼肌和肝脏AMPKα(Thr172)活性的连续激活可能是运动延缓高脂大鼠IR形成的关键机制之一。

【Abstract】 Objectives: it is to observe the effects of exercise intervention during IR formation of high-fat-induced SD rats as well as some mechanisms.Methods: SD rats were divided into normal diet control group(C group),normal diet exercise group(E group),high fat diet control group(H group) and high fat diet exercise group(HE group).FINS,FBG,HOMA-IR,and ISI are used to evaluate the effect of exercise.And ELISA and real-time PCR are adopted to observe serum adiponectin,visceral fat tissue adiponectin mRNA expression.Western blotting is also used to determine AMPKa expression and its phosphorylation level of quadriceps muscle and liver.Results: 1) There is no significant difference of serum adiponectin in each group,while the visceral fat tissue adiponectin mRNA expression in H group is significantly lower than that in C group.2) There is no significant difference of quadriceps muscle AMPKa protein expression in each group,while the phosphorylation level of H group is significant higher than that in H group,and the level of E group is higher than in C group.3) There is no significant difference of liver AMPKa protein expression in each group,while the phosphorylation level of HE group is significantly higher than that in H group.Conclusions: The decrease of visceral fat tissue adiponectin mRNA expression may involve into the formation of IR in high-fat-induced rats.No significant effect of exercise has been found on serum adiponectin,visceral fat tissue adiponectin mRNA expression,showing an uncertainty of causal effect between exercise delaying on IR formation and the change of adiponectin after exercise.Exercise increases the AMPKA expression and its phosphorylation level of muscle and liver,showing the continuous activation of AMPKA may be the key mechanism of exercise delaying on IR formation in high-fat-induced rats.
