大孔树脂对芝麻粕中芝麻素的分离纯化

【摘要】 采用大孔树脂吸附法对芝麻粕中芝麻素进行分离纯化研究。静态吸附和解吸实验结果表明:AB-8和HP-20对芝麻素有较高的吸附率和解吸率,其吸附率分别为9.24mg/g和9.28mg/g,解吸率分别为94.81%和94.35%。AB-8大孔树脂动态吸附与解吸实验结果表明:进样质量浓度越高吸附量越大;进样流速越小越有利于吸附;95%乙醇解吸效果较好;解吸液的合理使用量为10倍柱体积(BV);洗脱流速越小,解吸率越高;综合考虑进样流速与洗脱流速选1.0mL/min为宜。经AB-8大孔树脂纯化后,芝麻素纯度由醇提粗提物的12.88%提高至70.56%。

【Abstract】 This work reports the use of macroporous adsorption resin to separate and purify sesamin from sesame seed meal.The results of static adsorption and desorption experiments showed that AB-8 and HP-20 resins had adsorption rates of 9.24 mg/g and 9.28 mg/g and desorption rates of 94.81%and 94.35%,respectively.In dynamic adsorption and desorption experiments,higher adsorption rate of AB-8 resin for sesamin was obtained by increasing sample concentration,and lower sample loading flow rate was helpful for the adsorption of sesamin.In addition,95%ethanol had good desorption efficiency and the best desorption results were obtained using 10 BV of 95%ethanol.The lower the sample loading flow rate,the higher the desorption rate.Together,our findings indicated that the optimum sample loading flow rate and desorption flow rate were both 1.0 mL/min.The sesamin content(i.e.,purity) of crude ethanol extract from sesame seed meal was 12.88% and increased to 70.56%after purification with AB-8 resin.
