胰腺导管腺癌患者血清中microRNA-155的表达及其临床意义

【摘要】 目的检测microRNA-155(miR-155)在胰腺导管腺癌(pancreatic ductal adenocarcinoma,PDAC)组织中、患者血清中的含量,探讨其作为PDAC血清学诊断标志物的可能性。方法应用荧光实时定量PCR法检测miR-155在20对PDAC组织和癌旁组织、70例PDAC患者和40例健康人血清、10对荷人PDAC小鼠和对照组小鼠血清及6株PDAC细胞株和其培养上清中的含量。结果实验结果显示,PDAC组织中miR-155表达高于癌旁胰腺组织(P=0.005);并且miR-155在PDAC患者血清中表达量高于健康对照组(P=0.004);在荷人PDAC小鼠血清中miR-155含量高于对照组小鼠(P=0.012);同时,6株PDAC细胞株miR-155的表达存在差异(P=0.000),miR-155在细胞内和在其培养上清中的表达具有高度一致性(r=0.628,P=0.005)。结论 miR-155在PDAC组织和患者血清中表达上调,提示其可能是PDAC血清学的潜在标志物。

【Abstract】 Purpose To investigate the expression of microRNA-155(miR-155) in pancreatic ductal adenocarcinoma(PDAC) tissue and serum,as well as diagnostic value of miR-155 in serum of PDAC.Methods Tissue specimens from PDAC and pancreas(n=20 and n=20,respectively),the sera of patients with PDAC and control group(n=70 and n=40,respectively),the sera of SCID mice with humans’ PDAC and control group(n=6 and n=6,respectively),the 6 PDAC cells and supernatant were analyzed via real-time RT-PCR for miR-155.Results The expression of miR-155 in PDAC tissue was significantly higher than that in pancreatic tissue(P=0.005).The expression of miR-155 in the sera of patients with PDAC was significantly higher than that in control group(P=0.004).The expression of miR-155 in the sera of mice with humans’ PDAC was significantly higher than that in control group(P=0.012).It was been found different expression level of miR-155 among 6 pancreatic cell lines(P=0.000),a positive correlation was observed between the expression of miR-155 in the 6 PDAC cells and supernatant(r=0.628,P=0.005).Conclusion The expression of miR-155 in the tissue and sera of patients with PDAC is significantly higher than that in control group.Therefore,serum miR-155 may be used as a potential biomarker in the diagnoses of PDAC.
