新型寡核苷酸对牙龈卟啉单胞菌感染成骨细胞增殖活性的影响
Effect of Novel Oligodeoxynucleotide on the Proliferation of Osteoblast Invaded by Porphyromonas Gingivalis
【摘要】 目的:通过检测在不同序列寡核苷酸(oligodeoxynucleotide,ODN)作用下,牙龈卟啉单胞菌(Porphyromonas gingivalis,Pg)感染的成骨细胞增殖活性的变化,探讨ODN对Pg感染成骨细胞生物学性能的影响。方法:以感染复数为100∶1的Pg感染人成骨细胞MG63,加入12条不同序列的ODN(1mg/L),共孵育2h、4h、8h和12h,MTT比色法检测细胞增殖。结果:与PBS对照组相比,5条ODN对Pg感染的MG63细胞具有显著促增殖作用(P<0.05)。结论:特定序列ODN具有显著促Pg短期感染的MG63细胞增殖的作用,提示ODN对感染状态下的成骨细胞增殖这一生物学性能具有一定的影响作用。
【Abstract】 Objective:To explore oligodeoxynucleotide(ODN)biology function on osteoblast invaded by Porphyromonas gingivalis(Pg)through testing the effect of different ODN sequences(ODNs)on the proliferation of osteoblast invaded by Pg.Methods:MG63were incubated with Pg(MOI=100:l)and 12different ODNs(1μg/mL).MTT assay was used to detect the cell proliferation respectively after 2h,4h,8hand 12hcoculture.Results:The mean OD values of five ODNs groups were significantly higher than those in PBS control group and other ODNs groups(P< 0.05vs all of others).Conclusion:Specific ODNs can significantly promote the proliferation of MG63invaded by Pg in short-term.It shows that novel ODN has a certain influence on the proliferation of osteoblast invaded by Pg.
