α-亚麻酸对高糖下人红细胞抗氧化系统和膜的影响

【摘要】 目的探明α-亚麻酸(ALA)对高糖环境下人外周血红细胞膜结构的保护作用,并探讨这一功能是否与其改善高糖环境所诱导的氧化应激及增强抗氧化的状态有关。方法采用高效液相色谱法(HPLC)检测丙二醛(MDA)含量,液相色谱-质谱联用法(LC/MS)检测谷胱甘肽含量,流式细胞仪(FCM)检测膜的磷脂不对称性,超扫描电子显微镜(USEM)检测红细胞膜的形态。结果与50mM葡萄糖(Glu)组相比,50μM、100μM、200μM ALA组的MDA产生量均显著降低(P<0.01),GSH含量均显著升高(P<0.01),GSSG含量均显著降低(P<0.01),3组的红细胞形态也较50mM葡萄糖组正常。结论 ALA可能通过改善红细胞内氧化应激状态而发挥保护红细胞结构和功能的作用,进而可能对糖尿病并发症的形成起到预防作用。

【Abstract】 Objective To investigate whether α-linolenic acid could protect erythrocyte membrane structure in high glucose level and whether this effect could attenuate the oxidative stress induced by high glucose and improve the antioxidative capacity of erythrocytes.Methods The malondialdehyde and L-glutathione were detected by HPLC and LC/MS respectively.The membrane phospholipids asymmetry and fatty acid composition of membrane were demonstrated by FCM and GC respectively.The morphology of erythrocyte was illuminated by ultra scanning electron microscopy.Results It was found that:(1) the concentrations of MDA of three α-linolenic acid groups were significantly lower than that of the 50 mM glucose group(P<0.01);(2)the GSH level of three α-linolenic acid groups was significantly higher than that of the 50 mM glucose group(P<0.01);the GSSG levels of three α-linolenic acid groups were significantly lower than that of the 50 mM glucose group(P<0.01);(3) the labeled rates by FITC probe of three α-linolenic acid groups were significantly lower than that of the 50 mM glucose group(P<0.01);and(4) the erythrocyte shape of three α-linolenic acid groups and the PBS group was more normal that of the 50 mM glucose group.Conclusion ALA could protect the structure and function of erythrocytes,and then may have a preventive protection of diabetes and its complications.
