鸡IGFBP-3基因遗传多态性及其对京海黄鸡体重和产蛋量的遗传效应
Genetic variation at the MspⅠlocus in chicken IGFBP-3 gene and its associations with Jinghai yellow chicken body weight and egg production
【摘要】 采用PCR-RFLP技术和DNA测序技术对京海黄鸡、AA鸡、尤溪麻鸡、如皋草鸡4个鸡品种进行多态性检测,并将发现的SNP与京海黄鸡部分经济性状进行关联分析,研究4个鸡品种IGFBP-3基因第2外显子多态性及其对京海黄鸡生长和产蛋量的影响。结果表明:4个鸡品种中MspⅠ酶切位点基因型分布趋势一致,AA鸡多态信息含量最低(0.27)。χ2适合性检验表明,除京海黄鸡外其余3个鸡品种MspⅠ酶切位点均处于Hardy-Weinberg平衡状态(P>0.05)。关联分析结果表明,京海黄鸡CT和TT基因型个体的300日龄体重显著大于CC基因型(P<0.05)。京海黄鸡TT基因型个体的300日龄和66周龄产蛋量分别为111.94和186.80个,显著高于CC基因型个体。由此初步推断MspⅠ位点对京海黄鸡生长和繁殖性能有一定的影响,TT型为增加产蛋量的优势基因型,生产上可作为种鸡选育的候选分子标记进行选种。
【Abstract】 To reveal the polymorphism of the IGFBP-3gene exon 2among Jinghai yellow chicken,AA chicken,Youxi chicken,and Rugao chicken were used,and its relationship with growth and egg production in Jinghai yellow chicken were studied.PCR-RFLP techniques and DNA sequencing technologies were used to detect the SNPs of IGFBP-3exon 2,and analyze the correlation of SNPs with parts economic important traits in Jinghai yellow chicken.The result demonstrated that the MspⅠloci had the same gene frequency trend in the four chicken breeds,and the polymorphism information content(PIC)with AA chicken was the lowest among the four chicken breeds(0.27).Theχ2 test revealed that the mutation of the MspⅠsites among the other three varieties of groups except the Jinghai yellow chicken were up to HardyWeinberg equilibrium(P>0.05).Correlation analysis showed that,the body weight at 300days of CTand TTgenotype were significantly greater than those of CCgenotype(P<0.05),the egg production of TTgenotype was significantly higher than those of CCgenotype at 300days and the 66weeks(P<0.05),and the egg numbers of TTgenotype were 111.94and 186.80,respectively.The MspⅠsite is likely to have a certain role in promoting the growth and reproductive performances,while the TTgenotype which is the advantages gene of egg-laying.It could be used as a candidate molecular marker for breeding.
