实验性自身免疫性脑脊髓炎大鼠脑神经干细胞的增殖
Study on proliferation of neural stem cells in rats with EAE
【摘要】 目的 通过建立实验性自身免疫性脑脊髓炎动物模型,检测EAE神经干细胞的增殖。方法 以豚鼠脑脊髓匀浆加完全弗氏佐剂作为抗原,给予Wistar大鼠四足垫皮内注射,建立EAE模型,对正常组、免疫后7 d组、EAE发病后3,7,14,21 d组,分别进行5-溴脱氧尿嘧啶(BrdU)免疫组织化学染色,以观察正常大鼠脑内BrdU、EAE免疫损伤后BrdU的变化规律。结果 在正常组大鼠,Brdu阳性细胞主要分布在室管膜下区(SVZ)及海马齿状回的颗粒层(SGZ);Brdu阳性细胞数在免疫后呈现先上升后下降的变化趋势,于发病后3 d达高峰,21 d恢复正常。结论 EAE可以诱导大鼠脑内神经干细胞的增殖。
【Abstract】 Objective To study the proliferation of NSCs in experimental autoimmune encephalo-myelitis(EAE)at different time points by establishing an EAE animal model.Methods Wistar rats got foot-pad intradermal injection with the emulsion homogenate contained spinal cord of guinea pig and complete Freund’s adjuvant.Immunohistochemical staining was performed to examine the expression of BrdU in the normal rat brain and the variation law of BrdU after immune injury at different time points including groups from the control,7th day after immune as well as the 3th day,7th day,14th day and21st day after onset of EAE.Results The Brdu-positive cells were mainly located in subventricular zone of lateral wall(SVZ) and subgranular zone of dentate gyrus(SGZ)in control group;BrdU expressed unequally in different time points.The Brdu-positive cells increased at first,reached a peak at the 3th day after onset,then decreased and back to normal level at the 21st day after onset.Conclusions Experimental autoimmune encephalomyelitis(EAE) can induce proliferation of neural stem cells in rats.
